Objective This study was carried out in order to explore new X-STR markers and their multiplex PCR assay to improve the discrimination power of the X chromosome for forensic applications.We selected three X-STRs,which were GATA165B12,DXS6800 and GATA31E08, and constructed in an analysis system with multiplex PCR design strategy.Method Firstly a total of 154 unrelated Chinese Han female and 65 unrelated Chinese Han male in HeNan Provice samples were tested with the X-STRs separately by PAGE with silver staining system. Secondly, we constructed in an analysis system with multiplex PCR design. The multiplex PCR products were detected on ABI PRISM 3100 Genetic Analyzer using four labeling colors. Lastly, a series of validation experiments were carried out according to the suggestions by the Technical Working Group DNA Analysis Methods (TWGDAM)。Result The three loci we studied here were STRs on the X chromosome, the number of alleles observed at these three X-STR loci was 4,7,and 12, heterozygosity was 0.617,0.266 and 0.766 respectively. DP of the X-STR GATA165B12,DXS6800 and GATA31E08 was 0.806,0.46 and 0.906, PIC was 0.57,0.28 and 0.72, PE was 0.312,0.051 and 0.538, TDP was 0.99015, CPE was 0.6984 in 154 unrelated Chinese Han female in HeNan Provice samples respectively. The number of alleles observed, yielded 28 haplotypes in 65 unrelated Chinese Han male in HeNan Provice samples.Conclusion Our study reveals that the three X-STR loci we studied are suitable candidate X specific markers for forensic application in our population. The multiplex system established is proved to be robust and reliable with both simulated and real casework samples. It can be applied for the forensic DNA typing.
