With the development of free radical biology & medicine, the relationship between free radical in vivo with many diseases and physiology gradually been recognized. It has a great significance that exploring the prevention, diagnosis and therapy of free radical damage in vivo from the free radical perspective. In this study, the acute stress injury mouse model induced by DMF was investigated. The changes of the ascorbic acid and ascorbyl radical in vivo were measured by ESR, in order to establish a biomarker for diagnosing oxidative stress induced by DMF. This is very important for the research of occupation disease induced by DMF. In addition, the exogenous antioxidants which can prevent and remedy the radical damage were detected. UV spectrometry, electron spin resonance and chemical luminescent methods were used to measure the antioxidant activities of eleven kinds of common Chinese herbal medicine.2,2-diphenyl-l-picryl-hydrazyl radical(DPPH-),hydroxyl radical(·OH) and superoxide radical(·O2-) were detected respectively for making a comprehensive evaluation. The results indicated that majority herbs have good antioxidant activity. And the lilac, old crane grass, rhubarb have the strongest antioxidant activity. The findings also suggested that each herbal presents different scavenging abilities to different free radicals. For each free radical, the results detected by different methods also had some discrepancy.Contrast to electron paramagnetic resonance technique, there is much better consistency in measuring scavenge ability of antioxidant to active oxygen free radical by chemical luminescence.
