Background:ARHI and DAPK are autophagy-related tumor suppressor genes. DAPK plays a central role in autophagy,it can mediate membrane blebbing and the formation of autophagic vesicles. ARHI may play a role in presenting LC3 for DAPK-MAP1B. DAPK may play a role in gating in autophagic activity, which can provide the right conditions for ARHI. ARHI and DAPK may play a synergy role through regulating autophagic activity in carcinogenesis of endometrioid adenocarcinoma.Objective:This study is to assess the expression of ARHI and DAPK in endometrial carcinoma and to analyze the correlation of the expression level of ARHI and DAPK. At the same time, this study aims to correlate the expression with clinicopathologic parameters, which may discover the roles of ARHI and DAPK in the development of endometrial carcinoma.Materials and Methods:We collected 172 specimens of hysterectomia tissue (86 specimens of endometrial cancer,41 specimens of endometrial hyperplasia and 45 specimens of normal proliferative phase of endometrium which had been hysterectomized because of hysteromyoma). The expressions of ARHI and DAPK were detected by immunohistochemistry.SPSS 16.0 package was used for statistical analysis.Results:The expressions of ARHI in normal endometrium. endometrial hyperplasia and endometrial carcinoma tissues were100.00%,70.73%,50.00% respectively, the differences among the three groups were statistically significant (P<0.001). The expressions of DAPK in normal endometrium, endometrial hyperplasia and endometrial carcinoma tissues were 97.78%,75.61%,47.67% respectively, the differences among the three groups were statistically significant (P<0.001). The expression of ARHI was associated with cell differentiation,histological type,Surgical-pathological stage;The expression of DAPK was associated with cell differentiation and histological type.The expression of ARHI was positively correlated to that of DAPK in 86 specimens of endometrial carcinoma (R=0.763,P<0.001)Conclusions:1. ARHI and DAPK may play a role in carcinogenesis of endometrial adenocarcinoma.2:The low expression of ARHI and DAPK protein could help judging prognosis in endometrial carcinoma.3:ARHI and DAPK may co-participate in the carcinogenesis of endometrial adenocarcinoma.
