Expression Of IGFBP-3 In Endometrial Carcinoma Tissue

Objective: Endometrial cancer is one of the most common malignancy in Gynecology ,which its disease rate is the fourth of the female cancer, its pathogenesis has not been fully clarified. The Objective of our experiment is to investigate the expression of IGFBP-3 in endometrial carcinoma tissue and to explore its correlation with clinical behavior and pathological characters, as to investigate the relationship between IGFBP-3 and the development,infiltration,metastasis of endometrial cancer.Methods: Expression of Insulin-like growth factor binding protein-3(IGFBP-3) in 35 cases of patients with endometrial cancer and 29 cases with hysteromyoma were detected by Immunohistochemistry. The Expression of genes and their products of IGFBP-3 in the endometrial tissue were detected by real-time Q PCR. The serum levels of Insulin-like growth factor binding protein-3(IGFBP-3) in the patients were detected by Enzyme-linked Immunosorbent Assay(ELISA).Results: IHC:The expression of IGFBP-3 in endometrial cancer group was 31. 428%,over expression was 8.571%;expression of IGFBP-3 in control group was 93.103%,over expression was 55.172%。The rate of endometrial carcinoma was obviously lower. the immunostaining score was lower in endometrial cancer group than in control group(P<0.01)。In endometrial carcinoma, no relations was found between IGFBP-3 and histological type and tumor grade(P>0.05). The low expression rate of IGFBP-3 was significant difference between mymetria invasion, surgical stage and lymphnode matastasis (P<0.05).RT-PCR:The RNA levels of IGFBP-3mRNA in endometrial cancer group (0.633±0.479)were significantly lower than that of control group(1.273±1.405),The difference was statistical significance(P<0.05). In endometrial carcinoma, no relations was found between IGFBP-3mRNA and histological type and tumor grade(P>0.05). The low expression rate of IGFBP-3mRNA was significant difference between mymetria invasion, surgical stage and lymphnode matastasis (P<0.05).ELISA:In the serum,The expression levels of IGFBP-3(26.063±13.147μg/L) were significantly lower than that of control group(49.031±26.189μg/L), The differences were statistical significance(P<0.05)。In endometrial carcinoma, no relations was found between IGFBP-3mRNA and histological type and tumor grade(P>0.05). The low expression rate of IGFBP-3mRNA was significant difference between mymetria invasion, surgical stage and lymphnode matastasis (P<0.05).Conclusions: 1.The expression rate of IGFBP-3 in endometrial carcinoma was lower than that in normal endometrium. In endometrial carcinoma, the expression of IGFBP-3 are correlated with invasion and metastasis.2. These results demonstrated that The low expression levels of IGFBP-3 was associated with pathogenesis of endometrial cancer.
