Objective: to study the hazards of the nickel steel work to workers and the free radicals damageproductions are whether are as the occupational hazardous effect.Methods: To select steelmaking workers,steel rolling workers,steel slag disposingworkers in all 231 men as nickel exposure group,75 water pump workers as control groupby the different work-types. We investigated their general information- gender,age,thetotal work-age,the nickel exposure work-age,the occupational history,previous history,smoking,drinking and so on. We monitored the occupational hazardous factors of theproduction environment-soluble nickel compounds,dust,noise,hyperthermia by NationalOccupational Exposure Limits; to test the SOD activity by the xanthine oxidase method,theGSH-Px activity by the chemistry spectrophotometric method and the MDA content by thepenthiobarbital method; the urine nickel content tested by dimethylglyoximespectrophotometry; lymphocyte micronucleus rate detected by cytokine sis -blockmicronucleus-test; the urine 8-OH-dG content tested by ELISA; the repairase gene hGGO1mRNA expression tested by RT-PCR.Results: the highest nickel compound content was beside the steelmaking furnace,thehigher content was inside the steel slag disposing workshop,the minimal content was in thefront of rolling mill,no content detected in the water disposing plant; the SOD activity,theGSH-Px activity and MDA content of the nickel exposure group from the different work-typescompared to the control group, which had statistical significance(P<0.05); the SOD activity,the GSH-Px activity stepped down and MDA content increased with the work-age increasing, theSOD activity and the GSH-Px activity compared to the control group,0~group had no statisticalsignificance(P>0.05),the other groups had statistical significance(P<0.05), MDA content of thenickel exposure group from the different work-age compared to the control group, which hadstatistical significance(P<0.05). Micronucleus rate of different work-types compared tocontrol group had statistical significance(P<0.05) by Dunnett- T3 test, which had positivecorrelation with urine nickel content by correlation analysis(r=0.971); micronucleus ratesof different work- age compared to control group had statistical significance by Dunnett-T3 test, which had positive correlation with urine nickel content by correlation analysis(r=0.925); the urine nickel content of different work-types compared to the control group,which had statistical significance (P<0.05); the urine nickel content of different work-agecompared to the control group, 0~group and 6~group separately compared to the control group, which had no statistical significance (P>0.05),the other three groups compared tothe control group had statistical significance (P<0.05). 8-OH-dG content of the nickelexposure groups meanly was higher than of the control group, the steel rolling–worker groupcompared to the control group had no statistical significance (P >0.05),the other both groupsseparately compared to the control groups had statistical significance (P<0.05), hOGG1mRNA expression level of nickel exposure groups was higher than the control, which hadstatistical significance compared to the control group (P<0.05) and had positive correlationwith the urine 8-OH-dG content of different work-types by Pearson correlationanalysis(r=0.993) ; the 8-OH-dG content of different work-ages separately compared to thecontrol group had statistical significance (P<0.05), hOGG1 mRNA expression level of thedifferent work-age was higher than the control group, which had statistical significancecompared to the control group (P <0.05) and had positive correlation with the urine 8-OH-dGcontent of different work-age by Pearson correlation analysis(r=0.968).Conclusions: The occupational nickel exposure induced producing ROS, which made theoxidative damage; chromosomes and DNA were damaged by the free radicals, which presented thedocs-effect correlation, the damaged productions MDA,8-OH-dG were as the occupational nickelexposure markers.
