ObjectiveFor the model of rat's myocardial ischemia/reperfusion was established,to observe the effects of given the different density of Sevoflurane postconditioning on reperfusion injury after myocardial ischemia in rat. To investigate rat's myocardial protection of Sevoflurane on myocardial ischemia/reperfusion injury in rats and the best density.MethodsA total of 40 male healthy SD rats come form Shanxi medical university animal experiment center,200-250g body weight, were randomly divided into 5 groups:sham operation group (group C), ischemia/reperfusion group (group I/P),1% sevoflurane postconditioning group (group S1),2% sevoflurane postconditioning group (group S2),3% sevoflurane postconditioning group (group S3),8 in every group. The model of rat's myocardial ischemia/reperfusion was established.sevoflurane was inbreathed in 10 min after Myocardial ischemia for 40 min,After reperfused for 3h, blood sample was taken from carotid artery detecting the concentration of maleic dialdehyde (MDA) with the method of thibabituric acid (TBA),the activity of superoxide dismutase (SOD) with the method of xanthine oxidase and Creatine kinase-MB(CK-MB) Then killed the rat,The left ventricular specimens were obtained at the end of reperfusion.The morphologic changes of myocardium cell were observed under light microscope.Result1) Light microscopy:The results of hematoxylin eosin (HE) dyeing demonstrated that the model of acute myocardial infarction in mouse is builded successfully. The pathology pictures showed that,compared with I/P group, the degree of myocardial infarction in postconditioning groups relieved significantly.And compared with S1 and S3 group, the degree of myocardial infarction in S2 group relieved significantly.2) The level of MDA:The level of MDA in I/P,S1,S2 and S3 group were higher than that in group C(P<0.05).Compared with group I/P, the levels of MDA decreased significantly in group S1,S2 and S3(P<0.05). Group S2 is lower than group S, and S3 (P<0.05).3) The level of SOD:The level of SOD in I/P,S1,S2 and S3 group were lower than that in group C(P<0.05).Compared with group I/P, the levels of SOD increased significantly in group S1,S2 and S3(P<0.05). Group S2 is higher than group S1 and S3 (P<0.05).4) The level of CK-MB:The level of CK-MB in I/P,S1,S2 and S3 group were higher than that in group C(P<0.05).Compared with group I/P, the levels of CK-MB decreased significantly in group S1,S2 and S3(P<0.05). Group S2 is lower than group S1 and S3 (P<0.05).Conclusion1) Sevoflurane postconditionging can reduced infarct size and the level of CK-MB. Postconditioning with sevoflurane protects the heart of the rat against ischemia/reperfusion injury2) The protective effects made by sevoflurane were achieved by increasing the level of SOD and decreasing the level of MDA.3) Sevoflurane of different density could protects the heart of the rat against ischemia reperfusion in injury, and 2%sevoflurane has a better protective effect.
