Background and ObjectivesGastric carcinoma is one of the most common malignant tumors in China, which accounts for the leading cause of incident in the fourth place in all of malignant tumors, which accounts for the leading cause of death in the second place in all of malignant tumors, and which accounts for the first place in digestive tract cancer. Infiltration and transferation are the leading cause of death. Prompt diagnosis and prevention its transfer timely is one of difficult and hot spots in currenet research. Before transferation,cancer cell must get through the basement membrane many times. Therefore, the integrality of basement membrane and enzyme appear to the basement membrane degradation are vevy important.Claudins proteins are the main seal protein in tight junction. As main adhesion molecule between cells, the relationship of claudins and tumor transferation has attracted increasing attention.MMP-2 is one kind of matrix metalloproteinase. Matrix metalloproeines are a group of Fe2+ or Zn2+ dependence proteolytic enzymes, Now had proved that marix metalloproteinase play a key regulatory role in cancer's infiltration and transferation. Among matrix metalloproteinase, the relationship of MMP-2 and cancer's transferation is the most clostly. Gastric cancer cells and interstitial cells both can produce MMP-2, and the latter through the degradation of extracellular basement membrance to promote the transferation of gstric cancer.Under the situation of tumor iccidence rate increase year and year in our country, we hope that by detecting the Claudin-1 and MMP-2 proteins expression in gstric cancer, to find one new method to imporve cancer diagnosis and prognosis of gastric cancer, clinical better serveing in treatment.Materials and Methods1. Sixty-three cases of gastric cancer tissues samples were collectd, of which have fourty-eight cases of well-differentiated cancer, fifteen cases of poorly differentiated cancer, thirty-seven cases of positive lymph node metastasis, twenty-six cases of negative lymph node metastasis, Another twenty-seven cases of normal gastric tissue taken as control, After sample collection, they were embedded into paraffin section, and then they were made into tissue chip.2. The expression of Claudin-1 and MMP-2 porteins were spareately detected in gastric cancer tissues and normal tissues in the tissue microarray by the method of immunohistochemistry.Results1. In gastric cancer tissue the positive expression of Claudin-1 was 22, and the negetive expression was 41, and the positive expression rate was 34.9%, In normal tissues the positive expression of Claudin-1 was 19, and the negetive expression was 8, and the positive expression was 70.3%(χ2=9.576, P=0.002). Claudin-1 protein expression in lymph node-positive group was 8 cases, the positive rate was 21.62 percent; the expression of node-negative group was 18 cases, the positive rate was 69.23%(χ2=6.977, P=0.008). The positive expression of Claudin-1 was 20 in the well differntiated adenocarcinoma, the positive rate was 41.66%, and the positive expression of Claudin-1 was 2 in poorly differentiated adenocarcinoma, the positive rate was 13.33%(x2=4.037, P=0.045). The expression of Claudin-1 was not statistically significance in different gender and age groups.2. In gastric cancer tissues the positive expression of MMP-2 was 44, and the negetive expression was 19, and the positive expression rate was 69.8%, In normal tissues the positive expression of MMP-2 was 0, and the positive expression rate was 0(χ2=36.894, P<0.001), The positive expression of MMP-2 is 32 in the positive lymph node, and the positive expression rate was 84.68%, The positive expression of MMP-2 was 12 in the negetive lymph node, and the positive expression rate was 46.15%(χ2=11.793, P=0.001), The positive expression of MMP-2 was 33 in the well differentiated adenocarcinoma, the positive rate was 75.00%, and the positive expression of MMP-2 was 11 in poorly differentiated adenocarcinoma, the positive rate was 73.33%(χ2=0.114,P=0.736). The expression of MMP-2 was not statistically significance in different gender and age groups.3. Claudin-1 and MMP-2 had a negative correlation in gastric cancer (r=-0.679,P<0.01)Conclusions1. The protein of Claudin-1 can prevent to the infiltration and metastasis in gastric cancer.2. The protein of MMP-2 can promot to the infiltration and metastasis in gastric cancer.3. Claudin-1 and MMP-2 had played the role of different degrees in infiltration and metastasis in gastric cancer. Although with different mechanism, they express a certain degree of correlation between them. Claudin-1 with a low expression, MMP-2 with a high expression can be used as one high-risk indicator in infiltration and metastasis in gastric cancer. Through the detection of the two proteins have a certain significance to predict the prognosis of gastric cancer.
