Background and ObjectiveLung cancer is one of the most common primary malignant tumors, and its morbidity and mortality are all high. In china, the morbidity of lung cancer takes the first in various types of cancer, and it theatens the health and life of mankind seriously. For a malignant tumorits occurrence,development,infiltration and metastasis is procedure of several phases, multiple gene participation and several transcription factor regulating, and refer to a series of patho-physiological responses.The leading causes of mortality are infiltration and metastasis of lung cancer. So exploring the mechanism of the invasion and metastasis of lung cancer has become one of eseachhot spots.NGAL(neutrophilgelatinase-associated lipocalin) as a new member of lipocalin family was couducted and expressed in a variety of diseases, and is closely related to matrix metalloproteinase activity. NGAL was over-expression in the tumor cells that have the ability of metastasis. It is reported NGAL has something with breast cancer-free survival rate, but its expression and meaning in lung cancer is not known yet. Matrix metalloproteinase 10 (MMP-10) can degrade extracellular matrix, such as collagenⅥ, elastin, etc, and are closely related to tumor invasion and metastasis. But its expression and meaning in lung cancer is unclear.It has not been reported that the expression of NGAL and MMP-10 in lung cancer at home and abroad. In order to discuss the mechanisms of infiltration and metastasis of lung cancer and search valid methods which can suppress the infiltration and metastasis of lung cancer, this research adopts immunohistochemistry SP method to observe the expression of NGAL and MMP-10 in non-small cell lung cancer and normal lung tissues in side of cancer.Method1.Using the immunohistochemistry SP technology to detect the expression of NGAL,40 cases of lung squamous cell carcinoma tissues,30 cases of adenocarcinoma tissues and 30 cases of normal lung tissues in side of cancer.2.Compare these two biological markers of non small cell lung cancer with the clinical and pathological factors which influence the prognosis of lung cancer, analyze its use in lung cancer invasion and metastasis,and explore its ability to predict the biological indicators of lung cancer forecast.3.Statistical analysis:All the data were analyzed by SPSS 12.0 statistical package. The Chi-square was used in the comparison of positive rates, the t-test was used in the comparison of means between two groups,the One-Way ANOVA was used in the comparison of means between three groups, the relation of two variables was analyzed by the spearman level correlation analysis.The level of significant difference wasα=0.05.Results1.The positive expression rates of NGAL protein in lung squamous cell carcinomatissues, adenocarcinoma tissues were 62.5%,73.3%, The positive expression rates of MMP-10 in lung squamous cell carcinoma tissues, adenocarcinoma tissues were 65.0%,76.7%.2.The positive expression rates of NGAL protein and MMP-10 in lung cancer were 67.1%,70.0%.3.The positive expression rates of NGAL protein and MMP-10 in normal lung tissues in side of cancer were 20.0%,26.7%.4. The positive expression rate of NGAL protein and MMP-10 in the carcinoma tissues was compared with that in normal lung tissues in side of cancer from each other, and there was statistical significance (P<0.01). 5. Thus the expression of NGAL protein and MMP-10 in lung squamous cell carcinoma was compared with that adenocarcinoma, and there was no significance (P>0.05).6. The expression of NGAL protein significantly negatively correlated with TNM stages, grade of cell differentiation, lymph node metastasis, tumor diameter. The expression of MMP-10 significantly negatively correlated with grade of cell differentiation, lymph node metastasis, tumor diameter.7. Spearman correlation test show that the expression of NGAL protein and MMP-10 in lung cancer tissues were significant positive correlations.Conclusion1.NGAL and MMP-10 protein maybe participates in development of non-small cell lung cancer and also in infiltration and metastasis of lung cancer.2. NGAL protein and MMP-10 may play a synergistic effect during the process of development and infiltration, metastasis of non-small cell lung cancer.
