| Aim:To investigate the effect of Astragalosides (AST) and Panax Notoginseng Saponins (PNS) compatibility on the brain damage and the expression of p-JNK1/2,CytC,Caspase-9,Caspase-3 after cerebral ischemia-reperfusion, to probe into its Anti-ischemic brain injury protection mechanism. Methods: C57BL/6N mice were randomly divided into sham-operation group, model group, AST and PNS compatibility of high, medium and low-dose group, AST group, PNS group and Edaravone group. Cerebral ischemia-reperfusion injury were prepared by bilateral common carotid artery ligation for 20 min followed by 24 hours reperfusion after administration for 4 days. To detect pathomorphism with HE staining. to detect apoptosis with TUNEL ,the expression of p-JNK1/2,CytC,Caspase-9,Caspase-3 protein in brain was detected with western-blot. Results:The neurons was damaged,the neuronal survival rate was reduced and the neurons occurred apoptosis in the model group after ischemia-reperfusion for 24 and 48 hours. The neuronal survival rate in the drug groups was significantly higher than the control group(P<0.01), apoptosis rate was reduced (P<0.01) after treatment,and the effects of the-middle dose compatibility group and edaravone were more obvious, factorial analysis manifested that AST110 mg.kg-1 and PNS115 mg.kg-1 compatibility had a synergistic interaction (P<0.01). Western blot analysis showed that the expressions of p-JNK1/2,CytC,Caspase-9. Caspase-3 were increased notably in the model group after ischemia-reperfusion for 24 and 48 hours (P<0.01),and the expressions of p-JNK1/2,CytC,Caspase-9,Caspase-3 were lower in each drug group than the model group, the inhibitory effect of the-middle dose compatibility group on p-JNK1/2,CytC,Caspase-9,Caspase-3 was best, AST110 mg.kg-1 and PNS115 mg.kg-1 compatibility had a Superimposed interaction (P<0.05). Conclusion:1,AST110 mg.kg-1 and PNS115 mg.kg-1 compatibility had a synergistic effect against ischemia-reperfusion injury in mice,it related to anti-neuronal apoptosis after cerebral ischemia 2,AST110 mg.kg-1 and PNS115 mg.kg-1 has a synergistic antagonism on Neuronal apoptosis after cerebral ischemia-reperfusion, it May be related to inhibition of JNK signaling pathway activation,inhibition of mitochondrial apoptosis pathway. |
