Background and Objective:Glomerulosclerosis is a common morphologic change in almost all progressed renal diseases which characterized by progressive extracellular matrix accumulation. Traditional Chinese herbs have been found very effective to treat progressed renal diseases. though its mechanism is unclear now. To find the mechanism which traditional Chinese herbs treat Glomerulosclerosis, We established the models of Adriamycin-induced nephropathy and glomerulosclerosis in rats. At the same time, we investigated the effects of three kinds of traditional Chinese medicine prescription.Methods:120 Male Sprague-Dawley rats were divided into A, B, C, D, E, F and G groups at random. the rats of B, C, D, E, F and G groups were uninephrctomized and injected with adriamycin (ADR,5mg/kg body weight, except B group), by the first time 7 days later. to establish the models of Adriamycin-induced glomerulosclerosis, another dose (ADR,3mg/kg body weight except C group) were injected a month later. The E, F and G groups were given Jinkuishenqi tang, Jianpiyishen tang and Huoxuehuayu tang respectively 3 days before the second injection. Treated with traditional Chinese herbs for 2 months, all rats were sacrificed. Proteinuria was measured at preoperation, postoperation, interval of the 2 injection and the end of the study. Serum albumin, cholesterol, triglyceride,blood urea nitrogen and serum creatinine were determined before surgery and at the end of the study. renal histology was studied by morphometry; CollagenⅢ, collagenⅣand transforming growth factor-131 (TGF-β1) protein were evaluated by quantitative immunohistochemistry; collagenⅢm RNA expression were assessed by in situ hybridization. Analyse the data with SPSS12.0.Result:Rats of C and D groups showed FSGS which associated with tubulointerstitial injury consisting of tubular dilation and atrophy, interstitial leukocyte infiltration, and fibrosis. The kidney damage of D group was worse than the C group. The mRNA expression of collagen III was upregulated in diseased kidneys of C and D groups. The urinary excretion of protein progressively increased following the treatment with ADR, which was prevented partly by traditional Chinese herbs. Similarly, the glomerulopathy was also ameliorated by traditional Chinese herbs. Traditional Chinese may decrease the expression of collagenⅢ, collagenⅣand transforming growth factor-beta (1) which increased by ADR. However, jinkuishenqi tang could increase the death rate of adriamycin-induced glomerulosclerosis rats Which may be associated with the side effects of aconite. Jianpiyishentang, huoxuehuayutang could not prolong the life cycle of Adriamycin-induced glomerulosclerosis rats.Conclusion:It is suggested that the traditional Chinese herbal medicine can reduce proteinuria and retard the development of glomerulosclerosis. Perhaps the traditional Chinese herbal medicine exert its effect by inhabiting transforming growth factor-β1 (TGF-β1) protein expression which induced collagenⅢ, collagenⅣaccumulation in kidney. Among the three prescriptions, the effects of huoxuehuayu tang is the best, then the Jianpiyishen tang and Jinkuishenqi tang to cure Adriamycin-induced glomerulosclerosis in Rats. The traditional Chinese herbal medicine can't prolong the life cycle of Adriamycin-induced glomerulosclerosis rats.
