Chinese chives (Allium tuberosum Rottler. ex Spreng) is perennial herbs, belonging to the Liliaceae, which has special and pungent gas. It is a unique species of vegetables in our country, which has high productivity in Hebei province, Shanxi province, Jilin province, Jiangsu province, Shandong province, Anhui province, Henan province. Chinese chives have been considered some effects in blood circulation, hemostasia and stopping diarrhea, protecting liver and others. However, there are no studies or reports showed the effects of protecting livers from alcohol damage, assisting decreasing blood-lipid for hyperlipemia, antifatigue for kidney-yang deficiency and anticoagulation activity in vitro. So some biologic effects of Chinese chive were studied here by the model which alcohol leaded to mice's livers damage, the rats with hyperlipemia, the mice with kidney-yang deficiency and by anticoagulation activity in vitro, the paper want to verify these functions with crude extract of Chinese chives, then studied high activity function, for finding out which substance or compounds play the function.1. The experiment for protecting mice's liver by from Chinese chive was carried out. The mice's livers damage model were made, the mice were gavaged high dose alcohol (32mL/kg-bw,56°hong xing er guo tou). Crude extract of Chinese chive was gavaged with dose of 2.8850g/kg-bw for 7 days. The result showed that the crude extract maintained the mice liver's color obviously, and decreased AST and ALT activity in serum. When mice were gavaged middle dose alcohol (16mL/kg-bw,56°hong xing er guo tou) for 7days. The crude extract decreased AST and ALT activity in serum and liver. AST and ALT in serum decreased by 3.00% and 3.30%; AST and ALT in liver decreased by 4.20% and 6.10%. Chinese chive had slight effect for protecting liver damaged by alcohol to some extent. Refered to related literatures and compared to other liver-protecting tablet, Chinese chive only had less the function. It might be an assistant and could combined with other foods for protecting liver.2. Reduce hyperlipemia experiment of Chinese chive was carried out. Hyperlipemia model was made, the mice were given high-fat feed, crude extract of Chinese chive was gavaged with the dose of 2.8850g/kg-bw for 30 days. The result showed that the crude extracts decreased TC by 19.80%, the data was available; decreased TG by 11.80%, the data was unavailable; increased HDL-C by 0.028mmol/L, the data was unavailable. Refered to related standards, two kinds of available data were needed for applied decreasing blood-lipid function food. Chinese chive could play decreasing blood-lipid assistant. It could combine with other foods to reach to decrease blood-lipid auxiliary.3. Antifatigue experiment were carried out with Chinese chive, Kidney-yang deficiency model was made, the mice were intraperitoneal injected with hydrocortisone. The crude extract of Chinese chive was gavaged with dose of 2.8850g/kg-bw for 15 days. The result showed that the crude extract inhibited the rice weight reduce slightly, prolonged mice forced-swimming time by 3.22%. It decreased BUN by 3.22%, increased LDH by 0.70% in serum, and increased SDH in liver by 3.10% after mice had 30 forced-swimming minutes. Chinese chive had slightly antifatigue effect at the dose, we suggest Chinese chive need combine with related foods or increase dose to reach more antifatigue effect.4.10kg dry Chinese chive was extracted with 70% ethanol(v/v) at 60℃, then got the crude extract. The crude extract was extracted by petroleum ether, ethyl acetate, n-butanol and deionized-water, then concentrated in vacuum, got petroleum ether extract, ethyl acetate extract, n-butanol extract and deionized-water extract, after freeze-drying, we got petroleum ether extract 57lg, ethyl acetate extract 60g, n-butanol extract 36lg and deionized-water extract 1840g. The effect of Chinese chive experiment on anticoagulation activity in vitro were studied, n-butanol extract could decrease n-butanol anticoagulation activity obviously, the experiment showed n-butanol extract had promoting clotting substances; deionized-water extract had high anticoagulation activity.The deionized-water extract was precipitated by alcohol, the concentration of supernatant fractions and precipitate fractions were 0.0760g/mL for anticoagulation activity experiment in vitro, the experiment showed supernatant fraction and precipitate fraction both had anticoagulation activity, the supernatant fraction had higher anticoagulation activity than the precipitate fraction's.The supernatant fractions was eluted by a series rate deionized-water and ethanol on charcoal column. The results showed 95% ethanol eluting fraction had highest anticoagulation activity, and 95% ethanol eluting fraction's weight is 8.86g.95% ethanol eluting fractions was eluted by deionized-water on Sephadex LH-20,8 fractions were got, that was S1, S2, S3, S4, S5, S6, S7, S8. All treatment concentrations were 0.0760g/mL for anticoagulation activity experiment. The result showed S2 had highest anticoagulation activity, the effect of S1, S3, S5, S6 and S8 reduced in turn. When the rate of plasma to S2 was 20:1, the plasma didn't agglutinate, changed the rate to 40:1, the plasma agglutinated after 8.32 minutes. Color reaction for S fractions showed that the organic acid color reactions were positive; the steroid compounds color reactions were negative; S2 and other fractions showed blue fluorescence under 365nm. According to the color reaction results and literatures, the anticoagulant component of Chinese chive may be benzene propanoic acid components.
