ObjectivesTo explore the adjust mechanisms of innate immunity about certain Chinese herbs, and establish novel assay utilized in antivirus Chinese herbs.MethodsH1N1 influenza viruses infected cell model was constructed. The inhibiting effects on H1N1 influenza viruses from different concentrations of representative antivirus herbs components (OMT, I, APS, RD, GA, Ras) were observed by MTT colorimetry.The expression levels of antivirus immunity genes, i.e., IFNβ1, IL-29, IRF3 and ISG15, were examined by RT-PCR. Results were analyzed by the software of SPSS14.0.Results and DiscussionResults shown that the cell survival rate was 80.67% under the OMT concentration of 2mg/ml. And cell survival rates were 90.88%,80.84%,63.00%, 81.16%,68.64%, under the concentration of 375μg/mL(APS),5μg/mL(I), 1.25μg/mL(GA),2mg/mL(RD),250μg/mL (Ras), respectively.The virus proliferation restrain effects of Herbs components examined to H1N1 viruses infect Hela cells are obvious (P<O.01), some dosage depending relationship is shown between the medicine chroma and antivirus effect, there isn't obvious difference among vary chroma medicine antivirus effect.After each herbs act on cells, the expression of IFNβ1, IL-29, IFR are obvious ascended (P<O.05). the expression of ISG15 has little ascending (P>0.05), the ability of ascending the IFNβ1 expression is APS>GA>OMX>RD>Ras>I, the ability of ascending the IL-29 expression is RD>GA>APS>OMT>Ras>I>PolyI:C, there is obvious difference among RD, GA, OMT, APS(P<0.05). The ability of ascending the IRF3 expression among each medicine is not obvious, and there isn't obvious difference to gene expression by medicine chroma.There is not absolutely consistent gene expression with effect of herbs antivirus. The adjust inherence immunity system isn't only mechanism of herbs antivirus effect, herbs effect cell, research it gene expression can be a assistant means of herbs antivirus.Conclusion1. The studied Chinese herbs can regulation the expression of certain key antivirus immunity gene expression, such as IFNβ1, IL-29, ISG15 and IRF3.2. RD, APS, GA, I, OMT, Ras displyed marked effect on anti H1N1 virus.
