We have screened out the formula which contains Panax notoginseng (Burk.) F. H. Chen, Polygonatum sibiricumRed, Fritillaria cirrhosa D. Don and other 2 herbs, with the function of supporting the healthy energy, tonifying the qi and essence and nourishing lung yin, according to the literature search and pharmacology experiments. It applies to the symptoms:poor spirit, fatigue and lung yin deficiency and dryness caused by vital qi and nutrient substance deficiency. The formula was supposed to be developed as health-care food with the function of enhancing the immunity. The subject explored the extract process, shape process, quality standard, stability and preliminary function evalution.The extract and purification process was optimized by using the method of one-factor research, orthogonal test and comparative experiments. The extract ratio of polygona-polysaccharose was set as the indicator to compare three extract methods, and we obtained the best process:adding 8 times water than Polygonatum sibiricumRed., boiling for 2 time and 1 hour each, filtering through by 300 item, concentrating till the ratio of the herb and extract was 1:0.7, adding alcohol till the alcohol concentration was 80%, depositing, keeping the deposition in vacuum desiccator under the temperature of 70±2℃, and crashing the dry extract till it can pass through 120-item sieve. The extract ratio of three kinds of saponin(e) in Panax notoginseng was set as the indicator for optimizing the process by orthogonal test, based on the selected extract method and solvent.The best processing was:adding 12 times alcohol than Panax notoginseng coarse powder, regurgitating for 3 times,1.5 hours each time, vacuum concentrating under 77±2℃, vacuum drying for 12 hours under 77±2℃, and crashing the dry extract till it can pass through 120-item sieve. The Fritillaria cirrhosa was micronizated. And then the three powder was mixed even for use. One factor test was applied for selecting the granule accessories, while the briquettability, hydroscopic property, water solubilization and liquidity were set as index. We obtained the best formula and process:70% alcohol as cementing substance, pelletizing by 12-item sieve and vacuum drying for under 77±2℃, choosing the granule and doing package by holistic Scoring. Three groups of industrial products met the requirements, according to the quality examination.TLC was used to indentify Fritillaria cirrhosa, Panax notoginseng and Polygonatum sibiricumRed. respective, and specificity was good. HPLC was used to measure the content of notoginsenosideR1, Rg1, Rb1 and the method was accurate, sensitive and reliable. We have done accelerated stability test of 3 months, which proved all indexes were be up to the mustard. The animal immunity was proved to be enhanced, by doing the serum haemolysis measurement, antibody-producing cells measurement and DTH on low immunity mouse caused by cyclophosphamide.
