Remarkably, identification of effective and index components or the method of quantitative determination is not recorded in Chinese Pharmacopoeia (2005).By taking Radix Cyathulae ethanol extractives as the research object, Five compounds, cyasterone(Ⅰ), OH-cyasterone (Ⅱ), 3-O-{a-L-Rhamnopyranosyl-(1-3)-[β-D-glucopyranose(1-2)]-a-D-glucopyranosidurona te}-28-O-β-D-glucopyr anosyl oleanolic acid (Ⅲ) 3-0-[a-L-Rhamnopyranosyl-(1-3)-β-D-glucopyranosiduronate]-28-O-β-D-glucopyr anosyl oleanolic acid(Ⅳ), 3-0-[a-L-Rhamnopyranosyl-(1-3)-a-D-glucopyranosiduronate]-23-OH-28-O-β-D-glucopyr anosyl oleanolic acid(Ⅴ) were isolated from the 75% alcohol eluant with the macroporous adsorptive resin columns absorbing the Cyathula off icinalis Kuan, ethanol extractives. Through in-depth study, after ethanol extraction, macroporous resin purification, silica gel column chromatography, crystallization, drying, have high purity Cyathula reference substance cyasterone (HPLC determination of purity 98.8%).The Chinese Pharmacopoeia (2005), the quality specification of Cyathula off icinalis Kuan, only from the morphological and histological identification,but had not been given the method of TLC and content determination.In this study, based on previous research, developing an HPLC method for the content, determination of cyasterone in Cyathula off icinalis Kuan.,determining Cyathula off icinalis Kuan, of different processings, the content of cyasterone is 0.016%-0.101%, the proposed method is convenient,economical and accurate with reproducibility.This article has established quality standards of Cyathula off icinalis Kuan, by using modern analysis technology, to provide scientific basis of establishing quality standards of Cyathula off icinalis Kuan, in Chinese Pharmacopoeia (2010).
