The Expression And Clinical Significance Of Pi3k Signaling Pathway In Multiple Myeloma

Objective To investigate the expression of PI3K and its role in multiple myeloma.Methods Paients: 35 paients with MM (13 males and 22 females,median age 53 years,range from 24 to 73 years)were enrolled in this study from Mar.2008-Sep.2009.All paients were diagnosed according to the national criteria.The paients with bronchus asthma,renal diseases without MM,rheumatic diseases,other malignant tumor and serious infection were excluded.37 paients who were folled up were selected to detect PI3K level after finishing three chemotherapy courses.Controls:all 18 healthy controls,(9 males,9 femals,median age 51.1 years range from 41 to 65 years) were enrolled in this study. PI3K was quantized using standard enzyme-linked immunosorbent assay.All datas were performed using SPSS17.0 software.P<0.05 was considerd significant.Results (1)Overall, PI3K level were significantly higher in the 35 paients with MM when compared to normal controls (P=0.006<0.05). (2) Define the cases in the experimental group who were newly diagnosed or after treatment the disease still in advanced to be the invalid group, the rest were all involved in the effective group(the newly diagnosis patients are not involved in).PI3K level were significantly higher in the invalid group than the effective group.(P=0<0.05).(3)No significantly in the types of paients with MM.(4)PI3K level significantly correlated with the presence of bone lesions(r=0.547,P=0.001<0.05).A significantly larger proportion of MM paients with bone 3 grade had increased level of PI3K compared to paients with MM with bone 1 and 2 grade.(5) PI3K level significantly correlated with the DS stages (r=0.541,P=0.002<0.05). PI3K level were significantly higher in the paients with stageâ…¢a,â…¢b MM when compared to stageâ… -â…¡MM paients. (P1=0.003,P2=0.004<0.05),and there is no deference between stageâ…¢a andâ…¢b .(6)PI3K level significantly correlated with the ISS stages (r=0.672,P=0<0.05).PI3K level were significantly higher in the paients with stage 3 MM when compared to stage1,2 MM paients.(P=0.018<0.05).(7)PI3K level significantly correlated with the performance statues(r=0.419,P=0.029<0.05).PI3K level were significa- ntly higher in the paients with stage3,4when compared to stage1,2 (P=0.029<0.05).(8)After treatment 4 times,23 cases were involved.15 validity(completely recovery+partial recovery+advancement),a significantly larger proportion of MM paients before treatment 4 times had increased level PI3K compared to paients with MM after treatment 4 times (P=0.003<0.01);8 invalidity, no significantly proportion of MM paients before treatment 4 times had increased level of PI3K compared to patients with MM after treatment 4 times (P=0.152>0.05).Conclusion (1)The level of PI3K significantly correlated with the degree of active disease, the degree of bone destruction ,clinic stages, performance status.(2)The level of PI3K in bone marrow plasma can effect curative efficiency, is a risk factor on curative efficiency.Therefore, PI3K is a factor which can forecast the state of the disease, estimate the prognostic.
