Objective1.To explore the effect of HGF on survival rate and angiogenesis of rat dorsal ultra-long random skin flap.2.To compare the biological activity of HGF and VEGF on angiogenesis of skin flap.Methods30 standard healthy adult S-D rats (offered by Experimental Animal Center of Guangxi Medical University),weighted ranging from 300g to 350g,male or female unlimited,were randomly coded and divided into three groups(group A,B and C) and with 10 in each. A stands for HGF group,B stands for saline group and C stands for VEGF group. After rat models of 8.0cm×2.0cm ultra- long dorsal random flap were estalished, group A were directly given subcutaneous injection with 50ng/ml HGF solution, group B were directly given subcutaneous injection with saline and group C were directly given subcutaneous injection with 50ng/ml VEGF solution. After 7 days of operation flap color,texture,capillary blood return,necrotic area and bleeding of cutting flap were observed and remaining survival area of flap was counted.Average capillaries density was calculated by 400 optical microscope after CD~+34 immuno- histochemical staining.The difference before and after operation and their respective difference with the saline control group were compared. ResultsFlap survival rate of group A,B and C was ( 82.85%±7.07%), (43.82%±12.03%), (72.83%±9.87%) respectively,SNK analysis of variance test for pairwise comparison shows the differences were statistically significant (P <0.05). Survival rate of group A was higher than group B and C,and group C higher than group B.Before the experiment average density of capillaries of group A,B and C was (12.70±1.35),(12.34±1.22),(12.95±1.03 ) respectively, SNK analysis of variance test for pairwise comparison shows the differences are not statistically significant (P >0.05). The results were (44.67±8.96), (21.60±4.30), (35.47±7.83) respectively after the experiment and SNK analysis of variance test for pairwise comparison shows that the differenceswere were statistically significant (P <0.05). A, B and C three groups before and after the experiment did matching t test respectively, three groups before and after the experiment was statistically significant difference (P < 0.05), the results before the experiment were higher than prior to the experiment.Conclusions1.HGF can promote the formation of vascular endothelial cells of skin flap and improve its survival rate.2.HGF has more powerful biological activity of angiogenesis than VEGF for skin flap.
