OBJECTIVESTo investigate the expression of hTERT protein and Livin protein′s in colorectal carcinoma tissues, adjacent cancer tissues and normal colorectal tissues, which will to help to explore the correlations between hTERT protein and Livin protein, and to discuss the relationship between the two proteins and the clinicopathological characteristics and the prognosis in colorectal carcinoma tissues.METHODSThe technique of immunohistochemistry assay is applied to detect hTERT protein and Livin protein′s expression in colorectal carcinoma tissues(n=178), adjacent cancer tissues(n=60) and normal colorectal tissues(n=30). For the statistical analyses,χ2 test or Fisher′s exact test is used for description of data, Life-Table and Kaplan-Meier methods are used for overall survival rate, and Log-Rank test is used for the significance test of effect of prognostic factors. Cox regression model is used as main statistical methods for prognostic factors analyase. All datas are analyazed using SPSS software version 16.0. RESULTS1. The positive expression rates of hTERT protein and Livin protein in colorectal carcinoma tissues and adjacent cancer tissues and normal colorectal tissues are 94.4%, 65.7% and 6.67%, 8.33% and 3.33%, 0, respectively. The positive expression rates of colorectal carcinoma are significantly higher than those in adjacent cancer tissues and normal colorectal tissues(P<0.05), but the difference between adjacent cancer tissues and normal colorectal tissues has no statistical significace(P>0.05).2. The positive expression rate of hTERT protein in histological grade is statistically significant difference between well differentiation and moderate differentiation(P<0.05); In different infiltration depth, there is statistically significant difference between T1+T2 and T3+T4(P<0.05); In Dukes stage, there is statistically significant difference between A and B, A and D, C and D(P<0.05); In different clinical stage, there is statistically significant difference betweenⅡandⅣ,ⅢandⅣ(P<0.05); In tumor metastasis, there is statistically significant difference between M0 and M1 (P<0.05).3. The positive expression rate of Livin protein in different infiltration dep -th is statistically significant difference between T1+T2 and T3+T4(P<0.05); In Dukes stage, there is statistically significant difference between A and B, A and D(P<0.05); In different clinical stage, there is statistically significant difference betweenⅡandⅣ,ⅢandⅣ(P<0.05); In tumor metastasis, there is statistically significant difference between M0 and M1 (P<0.05).4. The expressions of hTERT protein and Livin protein′s are consistent in colorectal carcinoma tissues. In our study, the positive expression rates of three different tissues show an increasing tendency. The expression of hTERT protein and Livin protein′s is positively correlated in colorectal carcinoma tissues by Spearman correlation coefficients(r=0.204,P<0.05) .5. Cox regression analysis demonstrated that different infiltration depth, tumor metastasis, lymph node metastasis, hTERT protein and Livin protein are independent risk factors, respectively, which may affect the prognostic. As hTERT and Livin protein expression stronger, the survival rate is degressive trend. When both of the hTERT protein and Livin protein are expressing, the prognosis of the patients will be worst.CONCLUSION1. hTERT protein and Livin protein′s expression has significant differ -ences in colorectal carcinoma tissue, adjacent cancer tissue and normal colo -rectal tissue, expression level of hTERT protein and Livin protein in colorectal carcinoma tissue is obviously higher than thos in adjacent cancer tissue and normal colorectal tissue respectively.2. The expression of hTERT in colorectal carcinoma tissue is significant correlated with histological grade, infiltration depth, clinical stage, Dukes stage, tumor metastasis; The expression of Livin in colorectal carcinoma tissue is significant correlated with infiltration depth, Dukes stage, clinical stage, tumor metastasis.3. The hTERT protein and Livin protein are involved in the process of colorectal carcinoma′s progress and metastasis. The expressions of hTERT pro -tein and Livin protein′s are consistent, their expression is positively correlated in colorectal carcinoma tissues. In colorectal carcinoma occurrence mechanism , they play a synergistic action.4. The hTERT and Livin protein may be the prognostic factors of colo -rectal carcinoma, to detect of the expression of them can be meaningful prog -nostic indicator in colorectal carcinoma patients.
