Objective: To investigate the difference on rs61150458 of SNPs(Single Nucleotide Polymorphism) in Nell-1 among prognathous mandible (study group) and normal mandible (control group),and to provide a theoretical basis about etiology of mandibular prognathism.Methods: According to the criterion, we collected 60 individuals with prognathous mandible (MP) and 45 individuals with normal mandible, diagnosed in the stomatological department of the first hospital affiliated ShanXi medical University during 2008-2010. 5mL venous blood was draw-off and the genomic DNA was isolated from the whole blood cell. The aim gene was amplified by polymerase chain reaction (PCR), and performed in polypropvlene gelatin after electrophoresing, then judged by contrasting with the Marker. Afterwards, Hinfâ… locating in rs61150458 acts on the aim gene, we can get the results after eledtophoresing. If appearing two scrolls in the gelatin, we can consider that this site was G; otherwise, appearing one scroll, this site was C. Finally, the datas of study group and control group were tested by SPSS 17.0 software package.Results: In study group, there were 10 individuals with one scroll of 217bp in gelatin which existed nearby 200bp;32 individuals with two scrolls which were 83bp and 134bp nearby 100bp;18 individuals with three scrolls which were 217bp,83bp and 134bp.In control group, 12 individuals with one scroll in gelatin were performed;23 individuals with two scrolls which were nearby 100bp appeared and 10 individuals had three scrolls. Finally, the gene frequencies of the study group and the control group had not statistic difference by X2 (P=0.514,P>0.05); the genotypes of two groups had not significantly different by X2 (P=0.400,P>0.05).Conclusion: The gene frequencies and the genotypes on this site rs61150458 of SNPs in Nell-1 had not statistic difference between the prognathous mandible and the normal mandible, so it could not be obtained that this site had a relation with the etiology of mandibular prognathism.
