| Objective: the content of isolated rat cardiac and skeletal muscle were detectde by immunoblotting (western blot) at high temperature (30℃) and low temperature (10℃) and to study the rule of the degradation of isolated rat cardiac and skeletal myoglobin ,and to discuss the relationship between the degradation of isolated rat cardiac and skeletal myoglobin and the time of death , in order to provide the scientific basis for forensic early time of death.Methods: 16 healthy SD rats, weighing 200g±50g, were randomly divided into high-temperature group and the hypothermia group, 8 in each group.Head of 16 rats were placed in a closed rubber gloves, and fixed with the palm of their limbs and trunk, until the oxygen suffocation, remove the myocardium and skeletal muscle, on the humidor, placed in the artificial climate chamber, a constant temperature of the high-temperature group is 30℃, the constant temperature of the hypothermia group was 10℃, humidity under constant 40%, and stored 0h, 12h, 24h, 36h, 48h, 60h, 72h after the extraction of rat tissue proteins,respectively.To research the degradation of isolated rat cardiac and skeletal myoglobin and the time of death, medical image combined with statistical analysis to anlysis the results, then,to observe the relationship between the degradation of isolated rat myohemoglobin and the time of death,.Finally, to reseach the difference between the rule of regulation of vitro rat cardiac and skeletal myoglobin in high temperature and low temperature condition.Results:1.On the high temperature and the low temperature conditions, the content of myohemoglobin in rats myocardial, skeletal myoglobin were downregulation with the extension of the time of death. 2. The degradation of cardiac myoglobin, skeletal myoglobin were faster in the high-temperature group compared with that in the hypothermia group. 3.On the high temperature and the low temperature conditions, the degradation of cardiac myoglobin in rats was slower than skeletal myoglobin .Conclusions: 1.On the high temperature and the low temperature conditions, the content of myohemoglobin in rats myocardial, skeletal muscle were showed regular change trend.May be here is a certein relationshipcan be inferred, it can be a index to deduce the death time .2,temperature affects vitro regulation of myoglobin.it is important to the reaserch of the impact of temperature to hte early death time.3,Western blot is a simple operation technique, and is suitable for forensic science research . |
