Objective:To research the protection of edaravone on lung of rats during ischemia-reperfusion,andexplore the possible mechanism。Methods:Fourty rats were fallsed into five groups randomly:①shame operation group (Group SO),operate on sternotomy only;②the ischemia/reperfusion group(group I/R),the left pulmonaryhills of rats were ligated for 45minutes and then reperfused for 120 minutes;③edaravone treatedgroups (GroupC1,Group C2 and Group C3 with different drug dosages) ,the left pulmonary hillsof rats were ligated for 45minutes and then were reperfused for 120 minutes and intravenousinjection of edaravone 5 minutes before ischemia. At the end of the experiment, the left lung washarvested and blood sample was collected. Blood MDA,lung tissue wet-to-dry(W/D) ratio,inducible nitric oxide synthase(iNOS) and Superoxide Dismutase(SOD)were measured,makinglung tissue section and observing the change in light microscope.Results:(1)Compared with SO group: GroupIR and edaravone group could markedly rise the MDAcontent and iNOS activity(P<0.05).Compared with groupIR,edaravone group can reduce theMDA content(P<0.01)while group C2 and group C3 could low iNOS activity(P<0.01).GroupC1had no significaned change(P>0.05).iNOS activity have significance between group C2 and andgroup C3(P>0.05).(2) Compared IR with shame operation(groupSO):TheW /D ratio were significantly higher.Although group C2 and C3 are higher than groupSO,but are lower evidently to group IR, groupC1 had no significant changes(P>0.05). Compared with groupIR,SOD is higher evidently ingroup edaravone, SOD activity have significance between group C2 and and group C3(P>0.05)(3) Pathological changes of lung tissues, alveoli structure holonomy ,the wall is smoothand thin, alveolar space had no exudation in group SO; interval of lung became thick and muchexudation in alveolar space,part atelectasis.Groups edaravone are alleviate it markablely to groupIR.Conclusion:edaravone has a protection on lung ischemia-reperfusion injury in rat lung warm ischemiamodel.It maybe through clearing oxygen free radical(OFR) and controlling effectively lipidperoxide,while it can alleviate ischemia -reperfusion cytotoxicity and reduce inflammatoryreaction by inhibiting iNOS expression.
