Objective: Through the inspection of Th17 cells , CD4+CD25HiCD127LoTreg cells and related cell factors level in peripheral blood of lung cancer patients , analyzes the relationships of Thl7 cells and Treg cells with clinical stage and pathological types in lung cancer patients,so that we can explore the relationships about Th17 cells and Treg cells with occurrence and development of lung cancer and whether there are imbalances of Treg /Thl7. Methods: Through flow cytometry analysis and enzyme linked immunosorbent assay (ELISA) testing the proportions of Th17 cells and CD4+CD25HiCD127LoTreg cells in CD4+T lymphocytes and the expression level for related cell factors such as IL - 17, IL - 6, TGF - beta in peripheral blood of 70 patients with primary lung cancer, 36 healthy volunteers . Statistical analysis was performed by using the software SPSS13.0.Results: 1. The proportions of Th17 cells in CD4+T lymphocytes about peripheral blood of lung cancer patients(3.47土1.31%)was obviously higher than those of normal controls(2.09土0.73%)with significant difference(P<0.001). The proportions of CD4+CD25HiCD127LoTreg cells in CD4+T lymphocytes about peripheral blood of lung cancer patients(8.05土2.27%)was obviously higher than those of normal controls(6.15土1.54%)(P<0.001). The ratio of Treg/Thl7of lung cancer patients(2.36土1.23) was obviously lower than that of normal controls(3.05土2.18)(P<0. 05).2.There were no statistical differences among different TNM stages about the proportions of Th17 cells in CD4+T lymphocytes of peripheral blood in lung cancer patients (2.90土1.35% vs3.27土1.21% vs 3.78土1.35%)(P>0.05). There were significant statistical differences among different TNM stages about the proportions of CD4+CD25HiCD127LoTreg cells in CD4+T lymphocytes of peripheral blood in lung cancer patient(s6.24土2.16% vs 7.58土1.94% vs 8.87土2.27%)(P<0.05). Among them, The lung cancer patients in stageⅣwere signifieantly higher than those in stageⅠ-Ⅱand stageⅢ(P<0.05). There were no statistical differences among different TNM stages about the ratio of Treg/Thl7(2.73土1.65 vs 2.36土1.15 vs 2.36土1.33)(P>0.05).3.There were no statistical differences among lung cancer patients with different pathological classification (16 squamous carcinomas patients , 31 adenocarcinoma patients , and 23 small cell lung cancer patients) about the proportions of Thl7 cells and CD4+CD25HiCD127LoTreg cells in CD4+T lymphocytes and the ratio of Treg/Thl7 (P > 0.05).4. The level of IL-17,TGF-beta and IL-6 in peripheral blood of lung cancer patients was significantly higher than that of healthy controls . 5.The proportions of Thl7 cells in CD4+T lymphocytes was positively correlated with those of CD4+CD25HiCD127LoTreg cells( r=0.24,P=0.048). Conclusion 1. Th17 cells and Treg cells increased significantly in lung cancer patients ,especially in advanced stage.They may be one of the reasons that lead to immuno suppression and promote oncogenesis.2. Although the proportions of Th17 cells and Treg cells in peripheral blood of lung cancer patients obviously increased and formed positive correlation, the ratio of Treg/Thl7 of lung cancer patients was obviously lower than that of normal controls.The results may imply that exists in Treg/Thl7 imbalance and the increasing of Th17 cells is relatively dominat.3.The ratio imbalance of Treg/Thl7 may be concerned with their cell factors.
