Objectiveâ‘ We researched the protection of sevoflurane postconditioning on myocardium against ischemia reperfusion injury of rats in vivo by detecting the apoptotic index and expressions of Bax and Bcl-2 protein.â‘¡We further compare the intervention effects of given sevoflurane at verified different concentration on rat's myocardial ischemia reperfusion injury.Methods 60 healthy Wistar male rats which were from Shanxi medical university physiology animal experiment center with 200-250g body-weight were randomly divided into 5 groups(n=12):sham operation group (P group), ischemia reperfusion injury group (IR group), 1% sevoflurane postconditioning group (S1 group), 2% sevoflurane postconditioning group (S2 group), 3% sevoflurane postconditioning group (S3 group). Myocardial IR injury sample was established by ligating LAD. In P group, LAD was thread without other treatment. In IR group, the heart was ligated for 45min followed by 3h reperfusion. In sevoflurane postconditioning groups, the hearts were respectively inhaled 1%, 2%, 3% sevoflurane 15min before reperfusion. The left ventricular specimens were obtained at the end of reperfusion. The morphologic changes of myocardium cell were observed under light microscope. The expressions of Bax and Bcl-2 protein were measured by immunohistochemistry. Meanwhile the apoptosis index was detected by TUNEL.Resultsâ‘ The result of light microscopy: In P group, myocardial cells were normal. Compared with P group, IR group showed severe injury. The myocardial cells were lighter in S group than IR group.â‘¡The result of myocardium Bax and Bcl-2 protein: Compared with P group, expressions of Bax and Bcl-2 protein in IR group and S groups were significantly higher( P ?0.05).Compared with IR group, expressions of Bax protein in S groups were significantly lower( P ?0.05),while expressions of Bcl-2 protein and Bcl-2/Bax ratio were significantly higher( P ?0.05).There were no significant differences between S1 group and S3 group( P ?0.05).Compared with S1 group and S3 group, expressions of Bax protein in S2 group were significantly lower( P ?0.05), while expressions of Bcl-2 protein and Bcl-2/Bax ratio were significantly higher( P ?0.05).â‘¢The result of the myocardium apoptosis index: Compared with P group, AI in IR group and S groups were significantly higher( P ?0.05).Compared with IR group, AI in S groups were significantly lower( P ?0.05).There were no significant differences between S groups( P ?0.05).Conclusionâ‘ Myocardial ischemia reperfusion can increase expression of Bax protein, as well as decreasing expression of Bcl-2 protein.â‘¡Sevoflurane postconditioning can protect myocardial IR injury in rats by increasing expressions of Bcl-2 protein and Bcl-2/Bax ratio, as well as decreasing expressions of Bax protein and AI.â‘¢Sevoflurane postconditioning groups of different concentrations showed a clear cardio protective effect, and 2% sevoflurane post-processing have a better effect than 1% and 3% groups.
