Objective:This paper discussed the role ofβ-catenin in endometrial adenocarcinoma,s occurrence, development, invasion, metastasis and the significance of determining prognostic through testing theβ-catenin proteins,nuclear and plasma expression, lumens rim express/corresponding department express loss, gradient expression, lack of expression,differentials expression and it,s relationship with clinicopathologic factors; the correlation betweenβ-catenin mRNA expression and the protein expression, the correlation betweenβ-catenin protein expression and ER, PR gradient expression in endometrial adenocarcinoma.Methods:The expression ofβ-catenin in 87 cases of endometrial adenocarcinoma and 58 cases of normal endometrial tissue was detected by GBI universal two-step Immunohistochemistry. The study had analyzed nuclear plasma expression, lumens rim express/corresponding department express loss, gradient expression, lack of expression,differentials expression and the relationships between these expressions and clinicopathologic factors of endometrial adenocarcinoma, the correlation betweenβ-catenin mRNA expression and the protein expression, and the relationships betweenβ-catenin and endometrial adenocarcinoma,s occurrence, development, invasion, metastasis. Theβ-catenin mRNA expression in 52 cases of endometrial carcinoma and 22 cases of normal endometrium was detected by in situ hybridization. The correlations betweenβ-catenin mRNA and the expression of protein in endometrial adenocarcinoma were analyzed. Results1. Nuclear and plasma expression: the positive rates ofβ-catenin nuclear and plasma expression in endometrial cancer and normal endometrium were 52.9% and 34.5%. There was statistical difference between the two groups (χ~2 =4.746,p=0.029);. The expression rates ofβ-catenin nuclear and plasma expression in the low and middle differentation ( 65.8% ) was significantly higher than high differentation(42.9%), in stageⅡ~Ⅳ(67.6%) was obviously higher than in stageⅠ(43.4%), in the group with lymph node metastasis(62.5%)was higher than without lymph node metastasis(23.7%), which had statistical difference(χ~2 =4.517,p=0.034;χ~2 =4.889,p=0.027;χ~2 =6.632,p=0.010) .2. Lumens rim express/corresponding department express loss: the positive rates ofβ-catenin lumens rim express/corresponding department express loss in endometrial cancer and normal endometrium were 36.8% and 15.5%. There was statistical difference between the two groups (χ~2 =7.759,p=0.005);. The expression rates ofβ-catenin lumens rim express in stageⅠ(45.3%) was obviously higher than in stageⅡ~Ⅳ(23.5%), in the group without lymph node metastasis(42.6%)was higher than with lymph node metastasis(15.8%), which had statistical difference(χ~2 =4.215,p=0.040;χ~2 =4.607,p=0.032) .3.lack of expression: the positive rates ofβ-catenin lack of expression in endometrial cancer and normal endometrium were 56.3% and 3.4%. There was statistical difference between the two groups (χ~2 =42.667,p=0.000);. The expression rates ofβ-catenin lack of expression in >52 years group(63.8%)was significantly higher than≤52 years group(41.4%) ,in the low and middle differentation (76.3%)was significantly higher than high differentation(40.8%), in the group with lymph node metastasis(78.9%)was higher than without lymph node metastasi(s50.0%), in myometrial invasion≥1/2 (74.2%)was obviously higher than in myometrial invasion<1/2 (46.4%),in the group with squamous metaplasia (76.2%)was higher than in the group without squamous metaplasi(a50.0%), which had statistical difference(χ~2 =3.948,p=0.047;χ~2 =10.964,p=0.001;χ~2 =5.058, p=0.025;χ~2 =6.253,p=0.012;χ~2 =4.442,p=0.035) .4. gradient expression: the positive rates ofβ-catenin nuclear gradient expression in endometrial cancer and normal endometrium were 26.4% and 12.1%. There was statistical difference between the two groups (χ~2 =4.378,p=0.036);. The expression rates ofβ-catenin gradient expression in the group without risk factors(38.2%)was significantly higher than in the group with risk factors(18.9%), in stageⅠ(34.0%)was obviously higher than in stageⅡ~Ⅳ(14.7%), in myometrial invasion<1/2(33.9%)was obviously higher than in myometrial invasion≥1/2 (12.9%) which had statistical difference(χ~2 =3.995,p=0.046;χ~2 =3.949,p=0.047;χ~2 =4.536,p=0.033) .5. differentials expression: the positive rates ofβ-catenin differentials expression in endometrial cancer and normal endometrium were 67.8% and 15.5%. There was statistical difference between the two groups (χ~2 =38.221,p=0.000);The expression rates ofβ-catenin differentials expression in >52 years group(75.9%)was significantly higher than≤52 years group(51.7%),in stageⅡ~Ⅳ(82.4%)was obviously higher than in stageⅠ(58.5%), in the group with lymph node metastasis(89.5%)was higher than without lymph node metastasis(61.8%), in myometrial invasion≥1/2 (71.0%)was obviously higher than in myometrial invasion<1/2 (48.2%),in the group with squamous metaplasia (90.5%)was higher than in the group without squamous metaplasia(60.6%), which had statistical difference(χ~2 =5.161,p=0.023;χ~2 =5.404,p=0.020;χ~2 =5.224,p=0.022;χ~2 =4.199,p=0.040;χ~2 =6.512,p=0.011) .6. the expression ofβ-catenin mRNA: the positive rates ofβ-catenin mRNA in endometrial cancer and normal endometrium were 76.9% and 36.4%. There was statistical difference between the two groups (χ~2=11.159 ,P=0.001). In endometrial carcinoma, the expression ofβ-catenin mRNA was positively correlated with nuclear and plasma expression ofβ-catenin protein ,while was negatively correlated with lack expression ofβ-catenin protein(r =0.293, p=0.035;r =-0.284,p =0.041).7. The correlation between ER, PR gradient expression andβ-catenin mRNA expression: ER, PR gradient expression were no correlation with lumens rim express/corresponding department express loss ofβ-catenin protein(r =-0.066, p=0.542;r =-0.108, p =0.317), while were positively correlated with gradient expression ofβ-catenin protein ,negatively correlated with lack expression and differentials expression ofβ-catenin protein(r =-0.286,p=0.007;r =-0.248, p =0.020 ;r =-0.315, p=0.003;r =-0.320, p =0.002).Conclusion:1. Nuclear and plasma expression ofβ-catenin protein could play an important role in the occurrence, development of endometrial adenocarcinoma and in the process of its invasion and metastasis.2. Lumens rim express/corresponding department express loss and gradient expression ofβ-catenin protein may be biology indexes which prompt that the endometrial adenocarcinoma has low degree of malignancy and better prognosis.3. Lack of expression and differentials expression ofβ-catenin protein may be biology indexes which prompt that the endometrial adenocarcinoma has hight degree of malignancy and strong invasive and worse prognosis.4. The expression ofβ-catenin mRNA was negatively correlated with lack expression ofβ-catenin protein and positively correlated with nuclear and plasma expression ofβ-catenin protein,which could reflects a positive protective function which increase adhesion of tumor cell and stop migration and transfer of tumor cell.5. The gradient expression ofβ-catenin,ER and PR was a biology index which prompt that the endometrial adenocarcinoma has low degree of malignancy and better prognosis.
