Inhaled anesthetics has been used about 150 years old,but Anesthesia mechanism such as general anesthesia sites and the relationship between related molecules has not clarified for the complexity of the central nervous system,the uncertainty of related theory and the limitations of traditional research tools.Sevoflurane is widely used in clinical,it is a new inhaled anesthetic following enflurane and isoflurane,so this experiment choose sevoflurane as a representative of inhaled anesthetics.C- jun is one members of immediate early genes,it plays a very important role in signal transduction in nervous system. It can coupling the short-term afferent signal with long-term gene expression,change neurons plasticity and excitability,participate in neural cell growth,differentiation, information transmission,and other important life activities.a previous study has found the relevance among large receptor,molecules and general anesthesia mechanism,but the results disperse and lack of specificity.C-jun change quickly, can organic series and integration large signal.This experiment studies the effects of sevoflurane anesthesia on the levels of c-jun in the brain,provide reliable data and information,help to elucidate the anesthetic mechanism.Object:This experimental observe c-jun expression in some parts and related nuclei of the brain in mice by immunohistochemistry,analyze its possible reasons,explore the possible targets of sevoflurane,provide a new thinking for sevoflurane anesthesia mechanism.Methods:Male mice were randomly divided into four groups:group M-1,M-2,M-3 and M-4. Mice in the control group (M-1) were sacrificed by cervical dislocation after 5min oxygen uptake;group M-2 and M-3 undergoing sevoflurane anesthesia for 5min and 1h respectively;group M-4 receiving sevoflurane anesthesia for 1h and emerged from anesthesia for 1h. At the end of treatments,Mice are open-chest after dislocaitons executed, perfusion saline 100ml by heart,wash away the whole body blood,perfusion 4℃4% formaldehyde liquid 250ml.Remove brains while perfusion finished,fixed 90 minutes in the same fixed liquid,take the whole brain into 4℃refrigerator preservation in polyphosphate formaldehyde liquid.Specimens paraffin embedding,continuous coronary slice,tablet thickness 4um,HE dyeing,then immunohistochemical study.Using streptomycin resistance to biological grain protein-peroxidase method detecte c-jun antibodies.Extraction two slices randomly every place,one do immunohistochemical,compared with another one.The slice was placed under 40 times telescope,select five consecutive vision randomly,values dyeing strength said by average gray values.Results:The areas where c-jun positived cells changed obviously in the process of sevoflurane anesthesia,presented dissimilarity expression trend in group M-l,group M-2, M-3and group M-4.Concluded to the following 3 kinds:(1) C-jun positived cells expressioned in cell nucleus and cytoplasm of pallium,dorsal part (DMH),Edinger-Westphal nucleus (EW),lower expression and even without expression in group M-l,express significantly increased in group M-2,M-3.compared with M-2, expression no significant increase in DMH,The rest of the group increased significantly; c-jun positive neurons express weakened gradually or return to the control group level in group M-4.(2)C-jun positived cells expressioned in cell cytoplasm of periaqueductal gray (PAG),field CA1 of hippocampus,paraventricular thalamic nucleu (PV), ventral posterolateral thalamic nucleus (VPL),substantia nigra, medial part (SNM),anterior commissure,intrabulbar part(ACI),express significantly increased in group M-2,M-3 compared with group M-l.compared with M-2,expression no significant increase in CA1,The rest of the group increased significantly; c-jun positive neurons express weakened gradually,or express return to the control group level in group M-4.(3) C-jun positived cells expressioned in cytoplasm of lateral habenular nucleus (LHb),express significantly increased in group M-2,M-3 compared with group M-1.expression significant increase in group M-3 compared with M-2;c-jun positive neurons express weakened gradually,or express return to the control group level in group M-4.Conclusion:(1) C-jun positive neurons quantity in small brains are related to anesthesia-awakening process,c-jun maybe plays a certain effect in the general anesthesia different process. (2) Sevoflurane can induce the expression of c-jun positive neurons in pallium,DMH,EW,PAG,CA1,PV,VPL,SNM,AC1,LHB,these nuclear group may be sevoflurane role targets,may be involved in inhaled anesthesia mechanism. But the specific function under general anesthesia and mutual relation among related nuclear regiment still needs further study.
