The Effect Of Aldosterone On The Proliferation And Collagen Production And Related Genes Of Cardiac Fibroblasts

Objective To investigate the effects of aldosterone (ALD) on the proliferation of fetal cardiac fibroblasts (HFCFS) and the production of collagen I and collagenâ…¢in HFCFS,to seek the related seginal genes ALD induced. METHODS HFCFS were isolated by collagenaseâ…¡and then purified with differential attachment and detachment methods. The proliferation of HFCFS after ALD administration was assessed by CCK-8. The mRNA expression of COL1A1 and COL3A1 was assessed by reverse transcription polymerase chain reaction (RT-PCR). And the protein production of COL1A1 and COL3A1 was assessed by Western blot. Genechips of expression profile were used to detect different expressed genes.RESULTS ALD facilitated the proliferation of HFCFS concentration-dependently. ALD with lower concentration (10-9,10-8,10-7mol/L) significantly improved the expression of both COL1A1 and COL3A1,respectively.However ALD with higher concentration,such as more than 10-6mol/L,had no obvious effects on it. Genechips screeped out the different expressed genes such as CCL2,DAAM1,POLR2A,SDC1,HSPA1A,CLDN14.CONCLUSION ALD improved the proliferation of HFCFS concentration-dependently. And in a certain concentration range, ALD improved the expression of both COL1A1 and COL3A1,while with higher concentration beyond the range had opposite effects. On above process,there is no linear relationship between the effects of ALD on the proliferation of HFCFS, mRNA expression and protein production of COL1A1 and COL3A1.ALD could promote myocardial fibrosis and its signal pathways are complex.CCL2,MAKP,Wnt probably play a important role in the prosess.
