PartⅠ:The effects of Astragalus Polysaccharide on the cell proliferation, apoptosis, and insuline secretion of MIN6 cell linePurposes:To investigate the effects of different concentration of Astragalus Polysaccharide on the cell proliferation, apoptosis, and insuline secretion of MIN6 cell line.Materials and Methods:MIN6 cell line was cultured by different concentration of Astragalus Polysaccharide (0μg/ml, 10μg/ml, 100μg/ml and 1000μg/ml) for 48 hours. Cell proliferation was measured by MTT method, cell apoptosis by flow cytometry assay, and the function of insuline secretion was measured using different glucose concentration stimulation.Results:(1) The cell proliferation activity was higher in Astragalus Polysaccharide (100μg/ml) group than that in control group; (2) The cell apoptosis was lower in 10μg/ml and 100μg/ml Astragalus Polysaccharide groups than control group (P>0.05),while higher in 1000μg/ml Astragalus Polysaccharide group than that in control group (P<0.05); (3) The function of insulin secretion was higher in 10 and 100μg/ml Astragalus Polysaccharide group than that in control group (P<0.05),while lower in in 1000μg/ml Astragalus Polysaccharide group than that in control group (P>0.05).Conclusions:The MIN6 cell line showed increased cell proliferation activity, decreased apoptosis, and increased insuline secretion in Astragalus Polysaccharide with low and mediate concentration, The MIN6 cell treated by Astragalus Polysaccharide with high concentration was demonstrated decreased cell proliferation activity, incresed apoptosis, and decreased insuline secretion. PartⅡ:The effects of Astragalus Polysaccharide on the expression of P-AKT and PP2A activityPurposes:To investigate the effects of different concentrations of Astragalus Polysaccharide on the P-AKT expression and PP2A activity in MIN6 cell line.Materials and Methods:MIN6 cell line was cultured by different concentration of Astragalus Polysaccharide(0μg/ml,10μg/ml,100μg/ml and 1000μg/ml) for 48 hours. The expression of P-AKT was measured by Western blot method, PP2A activity by serine/threonine phosphatase assay system.Results:(1) Every group of the MIN6 cell line treated by Astragalus Polysaccharide showed increased expression of P-AKT, expecially in 100μg/ml Astragalus Polysaccharide group (P<0.05); (2) The MIN6 cell lines showed decreased PP2A acitivity in 10μg/ml,100μg/ml and 1000μg/ml Astragalus Polysaccharide groups (P<0.05).Conclusions:Astragalus Polysaccharide could increase P-AKT expression and decrease PP2A activity in MIN6 cell line.
