The thesis deals with the study on Picrasma quassioides (D.Dom)Benn., including the chemical constituents research, the HPLC fingerprint and quantitative analysis studies based on literature review. The aim is to establish quality standard and control the quality of this medical materials effectively.Chemical research of Picrasma quassioides (D.Dom)Benn.The isolation was with the intention of producing the alkaloids reference substances with high contents and strong pharmacological activities based on present literature review and preliminary HPLC-MS study on 85% ethanol extract of stem of Picrasma quassioides (D.Dom)Benn.. Nineteen compounds were obtained by integrated techniques including silica gel, etc. Nine ones of them were ascertained by physicochemical properties and modern spectroscopic analysis including UV,1H-NMR,13C-NMR, EI-MS and so on. They respectively are 4,5-Dimethoxycanthin-6-one (compound 3),1-Methoxycarbony-β-carboline (compound 5),3-Methylcanthin-5,6-dione (compound 6),3-Methylcanthin-5,6-dione (compound 7),β-Carbolin-1-yl 4,8-Dimethoxy-(3-carboline-l-lylethyl Ketone (compound 12),4-Methoxy-5- hydro-xylcanthin-6-one (compound 15),1-Propionyloxy-β-carboline (compound 16),1-Formyloxy-β-carboline (compound 18), 1-Formyloxy-7-hydroxy-β-carboline (compound 19). Compound(19) is a new natural product. In addition, compound(3), compound(5), compound(7), compound(13), compound(16) are active components according to literatures.The elucidated nine compounds consisted of P-Carboline type(four compounds), cathin-6-one type (four compounds)and dimmer type(one compound) alkaloids. Then they were subjected to HPLC-DAD-ESI-MS/MS to explore the fragment patterns. Their fragmentation pathway were found to be very similar, sharing the characteristic fragment ions of m/z 168, m/z 140, m/z 114. Furthermore, to establish a fast qualitative method for ethanol extract of Picrasma quassioides (D.Dom)Benn., extra fifteen constituents besides the former eight compounds were attributed to the chromatographic peaks, which could offer the foundation to the structure analyses of new compounds and chemical fingerprint study.Studies on quality control of Picrasma quassioides (D.Dom)Benn.The quality control researches on Picrasma quassioides (D.Dom)Benn. were promoted. The HPLC fingerprint method added was established on the basis of 31 samples of Picrasma quassioides (D.Dom)Benn from Guangdong, Guangxi, Jiangxi provinces. The fingerprint profile consisted of twenty-seven common peaks and fifty-six characteristic peaks, twenty and two constituents of them respectively were identified by chemical references and HPLC-DAD-ESI-MS/MS. The quality of 31 samples was analyzed by mathematical statistic methods such as of similarity and overlap ratio calculation, clustering and principle constituent analysis. The similarities of samples from Guangdong and Guangxi provinces were all over 0.820 showing no distinct difference between samples of the two producing areas, while the sample from Jiangxi province was below 0.770. The overlap ratio was low for those samples of low growth years. The result of clustering analysis was related to the total integral areas of common peaks of samples, and it reflected the quality difference of samples. The principle constituent analysis was based on the normalized data. The samples from Guangdong and Guangxi province could not be separated into two groups according to PC1-PC2 figure. And peaks of No.32, No.13 and No.23 achieved higher score in PC1 while peaks of No.50, No.5, No.32 scored higher in PC2.Moreover, simultaneous quantitative determination method of seven alkaloids (1-Formyloxy-7-hydroxy-β-carboline,1-Formyloxy-β-carboline,1-propionyloxy-β-carboline, 3-Methylcanthin-5,6-dione,4-Methoxy-5-hydroxycanthin-6-one,1-Methoxy-carbony-β-carboline,4,5-Dimethoxycanthin-6-one) was also established to assess the quality of Picrasma quassioides (D.Dom)Benn. from different habitats and medicinal parts and of different growth years. The results showed that there are significant differences between samples from Guangdong and Guangxi provinces and the alkaloids contents increase with the growth year. The stem alkaloids were higher than leaves and branches'.
