Banxia Xiexin Decoction and SiNi Powder are from the Zhongjing Zhang《Treatise on Exogenous Febrile Disease》,flat for cold and heat, Xiaopi Sanjie, look askance stomach weakness, cold and heat mixed, movements disorders, voicing confusion and Erzhi not stomach, abdominal pain, bloating, vomiting, diarrhea, In particular, the treatment of chronic gastritis. Semi-bionic extraction method is a whole drug molecule drug research and Combination, Pharmacy from the biological point of view, simulation of oral drug delivery and drug transit through the principle of the gastrointestinal tract, administration by the digestive tract for the design of Chinese medicine preparations, a new extraction process; CSG is a common disease of digestive system and frequently-occurring disease,but it is the lack of effective treatments in current.Objective:According to the principle of SBE, study on Banxia xiexin Decoction and Sini Powder extraction and prove its effections and mechanisms on treatmenting of chronic superficial gastritis, In order to discover the new therapeutic agents and methods for CSG,and develop the classical Chinese herbal formula also.Methods:By U9(91×33)uniformly designed and optimized extraction process on SBED, Pharmacodynamic targets and chemical indicators evaluating SBED, using a combination of factors to establish experimental chronic superficial gastritis (CSG) model:30% ethanol mixture with 2% sodium salicylate, three times a week; 10mmol·L-1 sodium deoxycholate, two times a week,; 0.05% ammonia as the freedom of daily drinking water consumption in rats; and ordered him to irregular diet, eat three square meals 2d, fasting 1d; and attach the clip end of anger, three times a week,2 hours each time. Modeling a continuous 42 days,42 days of modeling, the rats were randomly divided into 5 groups:Let the model control group, positive control group, SBED large dose group, SBED middle dose group, SBED low dose group. Model group was given normal saline 10ml·kg-1. Positive control group was given San Jiu Wei Tai suspension of 3.60g·kg-1 (equivalent to 10 times the clinical dose), SBED large, medium, low dose group were fed SBED solution 11.88,5.94,2.97g·kg-1 (respectively equivalent to the amount of clinical Medicinal 20,10,5 times),42 days continuouly. Animal behavior experiments, gastric mucosal morphology, determination of gastric secretion, gastric mucosal PGE2, NO, SOD, MDA determination, plasma PGE2, serum GAS, NO, MTL, SOD, MDA determination; Writhing induced by acetic acid to determine the analgesic effect SBED; to promote the rate of gastric emptying rate and small intestine of mice SBED index determination of gastrointestinal motility.Results:SBED extraction process for extracting the best temperature for 60±0.5℃, amount of water was 20 times the present meal weight 10,8,8 times, boiled water pH were 2.0,6.5,9.0; Minutes of boiling time were 140,70,35mins; the multi-factor alternative stimulus would make the CSG model successfully in 42-days, SBED can significantly improve the living condition and the pathological form of gastric mucosa to protect the shape and structure of gastric mucosal integrity on CSG; the gastric mucosal inflammatory cells, congestion and edema were reduced, mucosal thicken, glandular function recoveried; SBED can significantly improve gastric acidity, pepsin activity were increased, and the secretion of gastric juice were promoted, suggesting SBED can improve the digestive function; CSG models can be significantly increased serum gastrin, plasma motilin levels and lower levels; increase in serum PGE2 levels also, with a mucosal trophic effects; CSG models can be significantly increased serum and gastric mucosal SOD activity, decreased serum and gastric mucosal MDA, antioxidant, free radical scavenging effect; CSG models can be significantly increased gastric mucosal PGE2, NO content, has a protective effect on the gastric mucosa. SBED can significantly accelerate the intestinal propulsion and gastric emptying; SBED could significantly prolong the incubation period of writhing response in mice, can significantly reduce the acetic acid induced writhing in mice also.Conclusion:SBED can make the good therapeutic effect on CSG,improveing their living condition,forming of gastric mucosal pathology and improveing the stomach function on CSG, its mechanisms of action may be related to regulating gastrointestinal hormone, reducing free radicals and improving gastrointestinal motility.
