| Objective:In this study.we took SiHa cell line as working model,investigated the changes of proliferation ability and the effects of TSLC1mRNA expression on SiHa cell,after treatment with various concentrations of matrine,in order to further clarify the possible mechanism of inhibitory effects of matrine on cervical carcinoma.Methods:(1)The effect of matrine on proliferation of SiHa cell line measured by MTT assay Growing SiHa cells were affected by matrine at a final concentration of 0.5mg/ml,1.0mg/ml,1.5mg/ml.2.0 mg/ml,2.5mg/ml,respectively.After exposure to various concentration of matrine for 24h,48h,72h respectively.MTT was added and the plates were incubated for an additional 4h before absorbance at 490nm was measured.The inhibitory rate was calculated according to the formula.(2)Real-time PCR were used for detecting the relative expression of TSLClmRNA on SiHa cell line SiHa cells were collected after treated with 2.0mg/ml matrine or control medium for 24h,48h,72h,respectively.The total RNA were extracted from the cells and then Real-time PCR was performed to evaluate the TSLClmRNA expression.SiHa cells were collected after treated with 0.5mg/ml,1.0mg/ml,1.5mg/ml,2.0mg/ml,2.5mg/ml matrine respectively or control medium for 48h.The total RNA were extracted from the cells and then Real-time PCR was performed to evaluate the TSLClmRNA expression.Results:(1)The inhibitory effect of matrine on proliferation of SiHa cell line After 24h.matrine can remarkably inhibit the proliferation of SiHa cells with the concentration of 1.0mg/ml,1.5mg/ml,2.0mg/ml.2.5mg/ml(P<0.01).However,the inhibitory effect of matrine at the concentration of 0.5mg/ml was not significant(P>0.05).After 24h and 48h.matrine can remarkably inhibit the proliferation of SiHa cells with various concentrations of matrine(P<0.01).Therefore,in certain range of concentration.matrine can obviously inhibit the proliferation of SiHa cells in a dose-dependent and time-dependent manners.(2)The effects of matrine on TSLClmRNA expression of SiHa cell line After treated with 2.0mg/ml matrine for 24h.48h.72h.respectively.the relative expression of TSLClmRNA was 2.68±0.24.3.29±0.15,4.650.49.the TSLClmRNA expression of SiHa cell treated with 2.0mg/ml matrine of different time groups was significantly increased compared to that of control group(P<0.01).this effect was significantly different between various time groups(P<0.05 or P<0.01).Therefore,the effect of matrine had a time-dependent manners.After treated with 0.5mg/ml.1.0mg/ml,1.5mg/ml.2.0mg/ml.2.5mg/ml matrine for 48h respectively.the relative expression of TSLClmRNA was 1.52±0.76,1.94±0.67,2.48±0.29,3.29±0.15.4.35±0.43,the TSLClmRNA expression of SiHa cell treated with various concentrations of matrine for 48h was significantly increased compared to that of control group(P<0.01),this effect was significantly different between various concentration groups(P<0.05 or P<0.01).Therefore,the effect of matrine had a dose-dependent manners.Conclusions:(1)In certain range of concentration,matrine can remarkably inhibit the proliferation of SiHa cell line in a dose-dependent and time-dependent manners.(2)Matrine can up-regulate the relative expression of TSLClmRNA of SiHa cell line in a dose-dependent and time-dependent manners.(3)Matrine has a significant inhibitory effect on the proliferation of SiHa cell line by up-regulate the relative expression of TSLClmRNA. |
