| Object:Acute leukemia is a malignant cloning disease of hematopoietic stem cell.Because there are some limitations in various treatments, they are restricted in clinical application. In recent years, with the deep research of immune pathogenesis of leukemia, immunotherapy has been focused on leukemia therapy,especially on the areas of minimal residual disease, and has achieved considerable success. Newly discovered Th17 cells are an independent CD4+T cell subsets.Th17 cells and Treg cells regulate the immune stability process jointly. Their differentiation process is not independent, there is a certain contact between Th17 cells and Treg cells. The imbalance between Th17 cells and Treg cells is related to the occurrence and development of cancer and autoimmune diseases. This study aims to discuss the correlation between Treg cells and Th17 cells in acute leukemia and the changes of related cytokines(including IL-17,TGF-β,IL-6), describe the role of cellular immunity in acute leukemia, provide the basis for clinical immunotherapy.Methods:The expressions of Treg cells and Th17 cells were detected by Flow cytometry.The changes of related cytokines were detected by ELISA.Results:The ratio of Treg cells to CD4+ cells in acute leukemia group is highter than in control group, the differences were statistically significant, P<0.05. The ratio of Th17 cells to CD4+ cells in acute leukemia group is highter than in control group, the differences were statistically significant, P<0.05. It shows positive relation between Treg cells and Thl7 cells.The concentration of cytokines (IL-17,TGF-β,IL-6) in acute leukemia group are highter than in control group,the differences were statistically significant, P<0.05. The ratio of Treg cells to Th17 cells in acute leukemia group was lowter than in control group,the differences were not statistically significant,U>Uo.o5.Conclusions:In patients wiht acute leukemia,the number of Treg cells is increased,meanwhile,the number of Th17 cells is increased too.Treg cells and Th17 cells are postive relation.they may participate in the pathogenesis of acute leukemia.The density of cytokines(including IL-17,TGF-β,IL-6) is raised.These cytokines effect on the expression of Treg cells and Th17 cells,may be related to the occurrence of acute leukemia. |
