ADSCs (adipose-derived stromal cells) are being gotten more and more attention in different types of animal cells research. Although ADSCs have been obtained from experimental animals adipose tissue (eg. rats) and carried out in relevant studies, but few ADSCs studies have been reporteded on livestock (eg. goats).In this research a method of rat ADSCs isolation was used to isolate ADSCs from goat adipose tissue, Compared with rat ADSCs, goat ADSCs were to established and directed differentiated. The results showed that goat ADSCs cells had normal diploidy after subculture. Proliferation of goat ADSCs was faster than rats cells in the same conditions. Both rat and goat ADSCs stained positively for CD49d, CD44 and CD 13, but stained negatively for CD34 and CD 106. Bone nodules were apparent, and alizarin staining was positive after osteogenic induction. Cells expressing osteocalcin were positive by alkaline phosphatase (ALP) staining. After osteogenic induction, ossification nodules of goat ADSCs were larger than that of rat's with dense ALP staining. Adipogenic induction resulting in lipid droplets and peroxisome proliferator-activated receptor (PPARy2) expression were observed. Cartilage lacunae were formed and COL2A1 was expressed after cartilage differentiation. The differences that more cartilage lacunae with better morphology were observed following differentiation of rat ADSCs using the hang-drop method. For goat ADSCs, results of both adherent-induced and hang-drop induced cultures were better than that of three-dimensional cultures with more cartilage lacunae and better morphology. However, there was no significant difference of culture results between adherent and hanging drop induced culture.The results of this research indicated that a high purity goat and rat ADSCs had been established in this study.
