This paper studied on culture medium and fermentation end of submerged fermentation for Ganoderma lucidum, and components of different growth stages and different growth stages in different parts of G. lucidum fruit bodys.First, established a method to determine the content of triterpenoids in G. lucidum, and the methodology verification.The results showed that, circumfluence extract 3 times with absolute alcohol, and each time 2 h, can completly extracted triterpenoids in the samples.It could got good results that the products were edulcorated by saturated solution of NaHCO3.The content of triterpenoids was accurate determined by UV spectrophotometric method after glacial acetic acid-vanillin solution and high chlorine acid show color.Grown of G. lucidum to divided into Button stage,Stipe differentiation stage,Pileus differenti-ation stage,Mature fruit body stage,Spores mature stage, Spores released stage.To observemicr-oscopic characters and component difference of different stages by microscopical identification and thin layer chromatography(TLC).Contents of extract,hierarchical polysaccharides,teriterpen-oids,inorganic element,fatty acid at different stages were determined.Microscopic identification results showed that generative hyphae less and less,skelatal hyphae more, contact hyphae little change, whole growth process hyphae of clamp didn't always be found. TLC identification results showed that characteristic components have not changed at different stages. The result of determine the content showed that the contents of total polysaccharide and polysaccharides precipitated by 40% alcohol and 80% alcohol were the highest at button stage, the content of polysaccharide precipitated by 65% alcohol was the highest at mature fruit body stage. Whereas, the contents of triterpenoid, water-soluble extracts and alcohol-soluble extracts were highest at pileus differentiation periods. Content of K and AL followed by most Fe,Mg,Ca,and all kinds of inorganic elements tended to decrease as grow up of fruit body.29 fatty acids were identified, and confirmed the relative content by GC-MS.Kinds and relative content of fatty acids at different stages were different.8 fatty acids are found in Button stage,9 in Stipe differentiation stage,16 in Pileus differentiation stage,14 in Mature fruit body stage,24 in Spores mature stage,23 Spores released stage.9,12-octadecadienoic acid,cis-13-octadeceno-icacid,octadec-9-enoic acid,n-hexadecanioc acid had higher content. Based on principal component and reference other elements, preliminary conclusion that fruit body medicine for the purpose of the best harvest time is fruit body maturity period.G.lucidum fruit body was divided into the epidermis,tubes and stipe, the content of polysaccharides and triterpenoids of all parts at Mature fruit body stage, Spores mature stage and Spores released stage were determined.The results showed that, no matter which part is the highest content of polysaccharides in stipe, and the highest content of triterpenoids in tube. The content of polysaccharides and triterpenoids at each part of Mature fruit body stage have significant difference. Content of polysaccharides at each part of Spores mature stage have very significant differences.There were significant differences in the content of triterpenoids between epidermis and tube of Spores mature stage,and there were no differences between epidermis and stipe,tube and stipe. Spores released stage, there were no differences in the content of polysaccharides between epidermis and tube,tube and stipe.There were very significant difference in the content of triterpenoids between epidermis and tube,epidermis and stipe.There were significant difference in the content of triterpenoids between tube and stipe.the content of polysaccharides in epidermis and stipe has significant difference.The studied on effective ingredient content of different parts is to provide scientific for application of G. lu-cidum.The effect of carbon source,nitrogen source, inorganic salt on mycelial growth and active constituents accumulated was stuied by orthogonal test. The results show that, the influence of mycelial dry weight by sorts result set in descending order was inorganic salt, carbon sources, nitrogen source. The influence of polysaccharides and total active constituents by sorts result set in descending order was carbon sources, inorganic salt, nitrogen source. The influence of triterpenoids by sorts result set in descending order was inorganic salt, nitrogen source, carbon sources. Then a suitable culture medium of submerged fermentation for G. lucidum was obtained by comparing different production rates of total active constituents. Corn flour 4%, soybeans flour 2%, CaCl 0.225% and KH2PO4 0.15%, fermentation end for 120 h.
