【Objective】To evaluate the effect of gastric bypass on Goto-Kakizaki(GK) rat with type II diabetes mellitus with different fasting blood glucose level,and to find which level is the best choice in the fasting blood glucose level.【Methods】Type 2 diabetic rats were devided into four groups with different fasting blood glucose: GroupA(,≤10mmol/L,n=8); GroupB(,10.1~15.0mmol/L,n=8); GroupC,(15.1~20.0mmol/L,n=8); GroupD(,≥20.1mmol/L,n=8). All type 2 diabetic rats underwent duodenum bypass and 1/3 jejunum exclusion(gastric bypass,GBP) Rats were observed for 24 weeks after surgery. Fasting blood glucose (FBG) level,fasting insulin level,Hemoglobin A1c (HbA1c) level,food-intake and body weight were tested at 0, 1, 3, 6, 12, 24 wks. The 24wks descend ratio of glucose was calculated by formula.【Results】1. Fasting blood glucose (FBG) Blood glucose decreased after surgery in all the operative groups through the entire follow-up period (P<0.05). The glucose descend ratio of each groups was 32.6%,44.3%,40.3%,30.1%.and the level of 10~20mmol/L were the the best choice in the fasting blood glucose level2. Fasting insulin level Camparing to pre-operation, the insulin level of all groups was no significant change (P>0.05). Camparing to group A and B,group C and D have significant difference before and after surgery in all the operative groups through the entire follow-up period(P<0.05).3. Hemoglobin A1c (HbA1c) HbA1c of Groups of each time have no difference after operation (P>0.05) , But camparing to pre-operation, Fasting insulin level,Hemoglobin A1c level decreased after surgery in all the operative groups after operation of 12 wks (P<0.05).and have a stable level to each groups. 4. Food-intake and Weight Food-intake and weight have no difference before and after surgery in all the operative groups through the entire follow-up period(P>0.05).and blood glucose does not depend on the decline in food intake and body weight.【Conclusion】1. Roux-en-Y gastric bypass is best sect of the GK rat for fasting blood glucose control.2. Camparing to other level, These findings suggest that the more better effection is while the fasting blood glucose level between 10 mmol/L and 20mmol/L.This level is the best level.3. Blood glucose have difference after operation (P>0.05),But the improvement of blood glucose does not depend on the decline in food intake and body weight.
