Objective:To study the effect of pretreatment for rat free radical system of myocardial ischemia-reperfusion by injecting Xiangdan Zhusheye into PC6.Methods:The left anterior descene (LAD) coronary was ligated to establish the ischemia-reperfusion (I\R) heart model in the rats after fourteen days of pretreatment. The method of Colorimetric and Thibarbituric acid was to detect the content changes of XOD, SOD and MDA in blood serum and myocardium. Changes of histopathological in myocardium were also observed.Results:(1) The number and size of nucleus were reduced after ischemia-reperfusion. And it was increased on condition that the rats were pretreated by Xiangdan Zhusheye.Compared with compound Danshen Dripping Pills and injuecting normal sodium into PC6, the number and size of nucleus were increased by pretrated with Xiangdan Zhusheye.(2) As one of the sources of free radical generation, the content of XOD increased significantly, that means the growth factor of free radical in the upper reaches increased. Compared with Compound Danshen Dripping Pills and injuecting normal sodium into PC6, the content of XOD in blood serum and myocardium was increased by pretrated with Xiangdan Zhusheye.(3) As the clearing factor, the the content of SOD decreased significantly, that means the clearing factor of free radical decreased. Compared with injuecting normal sodium into PC6, the content of SOD in blood serum and myocardium was increased by pretrated with Xiangdan Zhusheye.The content of SOD in Danshen group and Xiangdan group was roughly epual, but the content of SOD in Xiangdan group shows a upward trend.(4) As the end product, the the content of MDA increased significantly, that means the end product of free radical increased. Compared with injuecting normal sodium into PC6, the content of MDA in blood serum and myocardium was decreased by pretrated with Xiangdan Zhusheye.The content of MDA in Danshen group and Xiangdan group was roughly epual, but the content of MDA in Xiangdan group shows a downward trend.Conclusion:Pretreatment for rat free radical system of myocardial ischemia-reperfusion by injecting Xiangdan Zhusheye into PC6 can decease the content of XOD and MDA, increase the content of SOD in blood serum and myocardium. Pretreatment by injecting Xiangdan Zhusheye into PC6 has significant protection to rat of myocardial ischemia-reperfusion through the way of intervenning the free radical system.
