| Objective: Colorectal cancer is one of the common malignant tumor, its incidence is increasing year by year. Its occurrence mechanism is still not clear. Currently believed to be caused by many factors such as diet , environment and heredity, and study confirms some disease is related with colorectal cancer incidence, for example: chronic inflammation, course longer ulcerative colitis formation granuloma. Colorectal adenomas is also prone to malignant transformation. Familial polyposis, the malignant transformation rate is almost 100%.Current research clear for diet and genetic factors, in the economic developed area, because eating structure to change, fat and protein intake more, cellulose intake reduced, it will lead to this area colorectal cancer incidence rate rise. There is a high risk in offspring of family history of cancer than those with normal. A variety of mechanisms involved in colorectal cancer occurrence, development and transferring, In numerous factors,a lot of research confirmed matrix metalloproteinases 9 (MMP-9) has been played an important role in tumors Occurrence, infiltration. and metastasis. At present, research about Filamin A (FLNa) and tumorigenesis, invasion and metastasis relation is seldom , especially in colorectal cancer.This study use immunohistochemistry methods to test FLNa and MMP-9,to explore relation between FLNa and MMP-9 and incidence of colorectal cancer and the relation between FLNa and MMP-9, and further to validate the relation in transcription level by using RT-PCR method, using Western imprinting method in protein level to verify it. Explore FLNa and MMP-9's relationship in colorectal cancer Occurrence, infiltration. and metastasis .Methods:1 Use immunohistochemistry method (SP method) to test the expression level of FLNa and MMP-9 in the normal bowel tissue and tumor tissue of colorectal cancer patients, make statistical analysis and then analyze the relation between the FLNa and MMP-9 in tumor tissue.2 Adopt RT-PCR method to detect the expression level of FLNa and MMP-9 in the normal bowel tissue and tumor tissue of colorectal cancer patients,and then make statistical analysis.3 Adopt Western imprinting method to detect the expression level of FLNa and MMP-9 in the normal bowel tissue and tumor tissue of colorectal cancer patients and then make statistical analysis.4 Use statistical analysis to analyze the relation between the expression level of FLNa and MMP-9 in tumor tissue and colorectal cancer patients age, sex, and tumor TNM classification,size, shape, infiltrating depth, location, bowel wall differentiated, lymph node metastasis, liver metastasis and histology clinical factors such as.Results:1 Immunohistochemical (SP method) detection MMP-9 and FLNa in the normal bowel tissue and tumor tissue of colorectal cancer patients results show that: the expression of MMP-9 positive particles in cytoplasm, in this experiment 60 patients with colorectal cancer positive expression is56 cases, the probability of 93.33%; By contrast in normal bowel tissue positive expression is 19 cases, the probability of 31.67% (.P<0.05, and the difference was statistically significant); FLNa positive particles is located in cytoplasm. in this experiment 60 patients with colorectal cancer positive expression is 30 cases,the probability of 50.00%; By contrast in normal bowel tissue positive expression is 55 cases, the probability of 91.67%. (P<0.05, the difference was statistically significant). Spearman test results confirmed: MMP-9 and FLNa in colorectal cancer adenocarcinoma tissue of expressing is negative correlation (r =-0.339, P = 0.008<0.05).2 RT-PCR method to detect MMP-9 and FLNa in colorectal cancer patients with the normal bowel tissue and tumor tissue results show that: the expression level of MMP-9 mRNA in the tumor tissue (0.84±0.02) is higher than the normal bowel tissue (0.21±0.01) (P<0.05, and the difference is statistically significant), the expression level of FLNa mRNA in the tumor tissue expression level (0.24±0.03)is lower than normal tissue (0.95±0.04) (P<0.05, and the difference is statistically significant).3 Western blotting method to detect the MMP-9 and FLNa in colorectal cancer patients with the normal bowel tissue and tumor tissue results show that: the expression level of MMP-9 protein in colorectal cancer tissue (0.86±0.03)is higher than normal tissue (0.09±0.02) (P<0.05,and the difference is statistically significant), the expression level of FLNa protein in colorectal cancer tissue (0.15±0.02) is lower than normal tissue (0.76±0.04) (P<0.05,the difference was statistically significant) .4 The relationship between the expression level of FLNa and MMP-9 and the patient clinical and pathological factors by statistical analysis shows that the tumor TNM classification, tumor invasion depth, lymph node metastasis intestines and liver metastasis has correlation, but the patient's age,gender, tumor location, differentiation degree, size,shape and histologic type has no correlationConclusion:1 The expression level of FLNa and MMP-9 has no correlation with the patient's age, gender, tumor location, differentiation degree, size, shape and type.2 The higher expression level MMP-9 ,the later TNM staging, the deeper bowel wall infiltration,the more lymph node metastasis ,and with liver metastasis.3 The higher expression level FLNa,the sooner TNM staging, the more shallow bowel wall infiltration,the less lymph node metastasis (or no) ,and without liver metastasis.4 In the test with FLNa and MMP-9,immunohistochemistry methods and RT-PCR and Western imprinting method has a consistent conclusion.5 The expression of FLNa and MMP-9 in colorectal cancer occurrence and transfer is negatively correlated. |
