Objective Discussion the expression of calmodulin-dependent protein kinaseⅡ(CaMKⅡ) on the hippocampus of rats Offspring with congenital HCMV infection. Provide scientific experimental evidence for congenital HCMV infection on learning and memory in rat offspring injury mechanisms for further research, and prevention of congenital HCMV infection induced learning and memory impairment and other neurological sequelae with disabilities.Methods See the bolt pregnant SPF SD rats were divided into experimental group and control group, n = 10. The HCMV virus with cytopathic effect (CPE) up to +++++++ was inoculated the 5th generation HELF cells. Cells degenerated to be more than 95%, In the 4 days of gestation, pregnant mice in experimental group were intraperitoneal injected of 0.5 mL HCMV suspension, and respectively, pregnant mice in control group were intraperitoneal injected the same amount of human embryonic fibroblast supernatant. The hippocampal neurons were prepared in 30 to 60 days rat offspring and the free calcium concentration ([ Ca2+]i) was fluorescence measured. Obtained the brain of rat Offspring and were observed pathology in hippocampus, and CaMKⅡof hippocampal for immunohistochemistry and Western blot experiments. The ultimate statistics was compared after that.Results Compared with the control group, in the cells band of hippocampus in rat offspring of experimental group under the light microscope, there was reduced number of cells, sparsely organization structure, disordered cells and degeneration disappeared neurons. Fluorescence results that the free calciumion concentration in hippocampus neurons of rats offspring with HCMV infection in early pregnancy in experimental group(153.47±37.26nmol/L) was significantly higher than the control group(102.32±21.33nmol/L), which had a significant difference (P<0.01). The immunohistochemistry positive products under light microscope were brown, and were mainly in the cytoplasm of pyramidal cells. The CaMKⅡimmunoreactive cells average optical density in rats offspring of experimental group (0.2139±0.1125) was significantly lower than the control group (0.2511±0.1097), which had a significant difference (P<0.05). The CaMKⅡaverage optical density of western blot in rats offspring of experimental group (0.4452±0.1351)was significantly lower than the control group(1.2558±0.1142), which had a significant difference (P<0.01).Conclusion Compared with the control group, the free calciumion concentration in hippocampus neurons of rats offspring with HCMV infection in early pregnancy increased in experimental group. HCMV infection in early pregnancy decreased the expression of CaMKⅡin hippocampus neurons membrane protein in rat offspring. These may be associated with damage on learning and memory function of hippocampus by congenital HCMV infection.
