Objective:The objective is to research the expression of WISP1 in the asthmatic model in rats Construction and the expression of it when RTED are stimulated by different kind of inflammatory factors. This paper, which considers Wnt signaling pathway as the research subjects, analyze the relationship of WISP1 and the morbidity of asthma, and on the basis of this, a further study on the molecular mechanism of the increasing expression of WISP1 in the asthmatic model will be done.Methods:Using ovalbumin (OVA) sensitized asthmatic model in rats Construction, determined the WISP-1 gene mRNA and the OD values of the lung tissue of rats by applying RT-PCR and Western blot. Used 9HTE cells, which were stimulated by IL-1β, IL-4, IL-13 and TNF-αfor 72 hours, to determine the WISP-1 gene mRNA and the OD values by the use of RT-PCR and ELISA.Results:1. Compared with the natural matched group, the WISP1 gene expression levels of asthma models of lung tissue have a significantly increased (P<0.05). It gradually increased with the course of time and reached its apex in the 12th week2. Compared with the natural matched group, after the stimulation of IL-1β,IL-4,IL-13 and TNF-αto airway epithelial cell, the mRNA level and the protein level of WISP1 greatly increased(P<0.05). the stimulation of TNF-αto airway epithelial cell is the most significant one for the mRNA level and protein level of WISP1 (P<0.01)。Conclusion:The expression of WISP1 increases in asthma models and it is involved in the development process of bronchial asthma. Its major function probably relate to the airway remodeling. The further study on the mechanism of WISP1 may present a new idea to treat the asthma airway remodeling. Under the condition of the asthma airway remodeling, the increasing expression of WISP1 has close relationship with TNF-α.And TNF-αtake part in the asthma airway remodeling by increasing the expression of WISP1.
