On the bases of this study, to observe the effect of M-Exo on proliferation, immigration, capillary-like tube formation in HUVECs, to explore the relationship between M-Exo and tumor angiogenesis.Methods1. The expression of EGFR in MDA-MB-231 were detected by immunocytochemistry and Western blotting.2. The preparation of M-Exo and EGFR detection: M-Exo were purified by serial ultracentrifugation and sugar density ultracentrifugation, The expression of EGFR in M-Exo were detected by Western blotting.3. To observe the effect of the M-Exo on EGFR expression in HUVECs: Co-cultured with HUVECs and 200μg/mL M-Exo for 24 h, the expression of EGFR in HUVECs were detected by immunocytochemistry and Western blotting, RT-PCR were used to detect the mRNA expression of EGFR in MDA-MB-231 and HUVECs after co-cultured with 200μg/mL M-Exo.4. To observe the effect of M-Exo on proliferation, immigration and capillary-like tube formation of HUVECs: MTT assay was used to detected the effect of M-Exo on proliferation of HUVECs. The change of HUVECs migration was detected by Transwell chamber method after treat with 200μg/mL M-Exo for 24 h. The capillary-like tube formations by HUVECs were observed.Results1. In the result of immunocytochemistry, the expression of EGFR in MDA-MB-231 were positive, the staining area were mainly brown granular in the cell membrane and cytoplasm. In the result of Western blotting, the positive reaction band was found in 170 KD which coincides with the molecular weight of EGFR protein.2. Western blotting result demonstrate that the expression in M-Exo were positive.3. Immunocytochemistry exhibited that part of HUVECs was found straw colour or brown madder particle in cytoplasm after treated with 200μg/mL M-Exo for 24 h , the ratio of positive expression was 21.4±3.1%. There were siginficant increase compared with control group(P<0.01). Western blotting showed that the positive reaction band was found in 170 KD which coincides with the molecular weight of EGFR protein. RT-PCR exhibited that the positive of EGFR mRNA in MDA-MB-231, and negative HUVECs after treated with 200μg/mL M-Exo for 24 h.4.MTT assay showed that the concentration range of M-Exo significantly increased HUVECs proliferation in a concentration-and time-dependent manner (P < 0.05). 200μg/mL M-Exo treatment group had the approximat best linear correlation. Then we chose the 200μg/mL M-Exo in our experiment. It may significantly enhance the migration of HUVECs after treated with 200μg/ml M-Exo for 24 h, The migration cell number of the control group and experimental group were 18.4±5.07 and 51.4±8.93, and the difference was significant compared with the control group(P<0.01).It may significantly promoted the capability of capillary-like tube formation of HUVECs, The number of tuber formation of control group and experimental group were 2.6±0.93 and 6.4±1.07 , and there were significant different compared with the control group(P < 0.05).Conclusion1. M-Exo with oncogene EGFR cargo, and could mediate EGFR protein to vessel endothelial cell. It may be the possible manner of abnormal expression EGFR in tumor vessel endothelial cell.2. M-Exo significantly increased HUVECs proliferation in a concentration-and time-dependent manner and it may promote the migration and the capability of capillary-like tube formation of HUVECs. There may be closely lincked between M-Exo and breast cancer angiogenesis. ObjectiveTo make know the current situation of health education on cancer and the way of obtain the knowledge of cancer prevention among Chongqing city.To explore the cancer health education strategy in the rural and urban residents of Chongqing, provide a theoretical basis for cancer prevention in Chongqing.Methods1. To survey the part of urban and rural residents in Chongqing. With self-designed questionnaire survey of urban and rural residents in Chongqing, to survey the current situation of cancer prevention education and the main way of access to the knowledge of prevent cancer.2.Respectively, for urban and rural areas to diversify forms of cancer prevention health education interventions, including lectures, prevention of cancer science image promotion, cancer prevention brochures and on-site consultation. 3. Another survey after the intervention of urban and rural residents in Chongqing Cancer knowledge and attitudes. Comparing the results of the survey after the intervention.The questionaire data were checked by double entry mesthod,using Epidata 3.1 software to establish a database and SPSS for statistical analysis.Results1.The overall level of health education to prevent cancer in Chongqqing city was low,and rural areas were lower than urban areas.2.The main ways of residents to access the knowledge to prevent cancer : mainly urban residents to television programs, followed by newspapers, magazines and Internet,mainly rural residents to television programs. On health education and promotion in the form of preferences is mainly lectures/expert advice.3. Urban and rural residents on the prevention of cancer low levels of knowledge of nurses,with poor health awareness and early symptoms of cancer awareness.4.After intervention by variety ways,residents of cancer prevention knowledge and malignancy rate of correct answers significantly inmpoved,and awareness of eaely symtoms of cancer prevention attitudes change significantly.ConclusionThe overall level of health education to prevent cancer in Chongqing city was low.
