Objective: To investigate the relationship between macrophages and hbv recurrence after liver transplantation with HBV-related patients.Methods: Collect 13 venous blood with anticoagulation from different HBV-related patients whose hepatitis B recurred after liver transplantation , while, we collect 14 examples from patients whose HBV is negative after liver transplantation to compare. Separate these examples though Ficoll-Hypaque density centrifugationicoll to get PBMC,then cultivate PBMC for 7 days with rhGM-CSF and RPMI-1640 which contained 10% calf serum to get adherent cell.Check out special antibody CD-14 with flow cytometryto assay the adherent cell,s nature. Detect HBV-CCCDNA within the cell,s though RT- PCR.Results: Of 13 patients who have HBV again after liver transplantation, 9 are HBV-CCCDNA positive within macrophages,the ratio is 69.23%. only 1 patient of 14 whose HBV do not recurrened ,it,s HBV- CCCDNA is positive,the ratio is 7.14% , Hepatitis B positive for HBV-CCCDNA relapse group was higher than that without recurrence group(P < 0.01). Conclusion: there probably be a relationship between HBV recurrence after liver transplantation and HBV CCCDNA lefted in their macrophages for those patients who have OLT.
