ObjectTo investigate the efficacy and mechanism of lysine acetylsalicylate (LAS) as aspirin complex salt in the treatment of lung injury caused by PQ poisoning.MethodThe rats were once intragastricly administrated with 50mg/kg PQ, and intervened with doses of LAS (200mg/kg). The content of malonaldehyde (MDA), the activities of superoxide dismutase (SOD), glutathion peroxidase (GSH-Px), and catalase (CAT) in serum and lung tissue, hydroxyproline (HYP) content in lung tissue, the pathological changes in lung tissue, the apoptosis rate and expression of Bcl-2/Bax protein in lung tissue, as well as the changes of T lymphocyte subsets were detected in each group on the 1st,3rd,7th,14th and 21st day.ResultsAfter intervention by LAS. the content of MDA in serum of the PQ+LAS group rats was lower than that in the PQ group on days 1,3.7and21 (all P< 0.05). the content of MDA in lung of the PQ+LAS group rats was lower than that in the PQ group on days 1.3 and 14 (all P< 0.05). Intervention with LAS could accelerate the recovery of SOD, GSH-Px and CAT activities in serum and lung, which showed the significantly differences on days 1,3 and 7 compared with the PQ group (all P<0.05). The HYP in lung of the PQ group rats began to increase obviously from the third day after PQ exposure (all P<0.05), and the treatment with LAS could also inhibited its procedure markedly (all P<0.05); After early LAS treatment, the apoptotic rate and Bax expression of lung decreased, the Bcl-2 expression increased, and the Bcl-2/Bax ratio increased, compared to those of the PQ group rats (all P<0.05); After intervention of LAS, CD3+, CD4+, and CD4+/CD8+ ratio in the peripheral blood were increased compared that of PQ group on days 1,3and7 (all P<0.05). after intervention of LAS, lung congestion, edema and infiltration of inflammatory cells at the early stage as well as the collagen fiber content at the later period were significantly relievedConclusionLAS had protective effects on injury of PQ posioned rats with through multiple channels, it could alleviate lung injury caused by PQ poisoning.
