| Chronic pancreatitis is one of the most common diseases of digestive system on clinical, and the incidence of the disease increases year by year.In recent years great progress has been made in the diagnosis and classification of chronic pancreatitis.However, the standards of classification are usually based on clinical manifestation,laboratory test and morphology and so on,the analysis of metabolism characteristics to the different classification of chronic pancreatitis has not been reported,and there is no uniform standard of classification to chronic pancreatitis on clinical.High-resolution Magic Angle Spinning Nuclear Magnetic Resonance Spectroscopy (HR-MAS-NMR) has been proved to be an effective method which can analysis metabolism ingredient in vitro without invastion,and it has high sensitivity in precisely analysing each part of compound in organization.In this reaearch,throurh to establish the different degree of chronic pancreatitis in rats,carry on HR-MAS-NMR with the vitro pancreatic tissues,and explore the metabolic characteriatic of rats with chronic pancreatitis from the normal as well as to define the correlation of the union pathology result and characteriatic metabolites,combining with the result of pathologic,the value of HR-MAS-NMR in the metabolic analysis and classification of chronic pancreatitis in rats will be explored.Objective : To explore the metabolic characteristics of chronic pancreatitis in an experimental model. Methods:With dibutyltin chloride (DBTC) solution (8mg/kg) injected via the tail vein, experimental model of chronic pancreatitis was constructed in 36 WISTAR rats. The 36 rats were divided into 6 groups, and each group consisted of 6 rats. On 0,7,14,21,28,35 days after modeling, rats in 6 groups was executed, pancreatic tissues of the rats was excised from the middle parts of each group.Each tissue sample was cut into two parts:one was used for histological test,the other was transferred into cygo-tubes,snap-frozen in liquid nitrogen and then storen at -80 prior to HR-MAS NMRS analysis.signal intensity of main metabolites were obtained from NMR spectra and statistical analysis was excuted with data.Results:Pathologic study had shown the rats of chronic pancreatitis can be divided into mild, moderate, and severe degree groups.and the severity of chronic pancreatitis increased gradually after modeling. Principal component analysis of HR-MAS NMR spectra showed that the Betaine (Bet) ,Phosphocholine and Glycerophosphocholine(PC/GPC),Choline (Cho)-contained components and 3-hydroxybutyrate(3HB) were significantly increased in severe chronic pancreatitis groups; Aspartate (Asp) , Lactate (Lac), Ileucine/Leucine/Valine (I/L/V) and Fatty acid (FA) were significantly decreased, when compared to mild chronic pancreatitis group and control group. The most significiant metabolites were Aspartate ,Betaine and Aspartate by various statistical analysis.Conclusion:HR-MAS NMRS was helpful in distinguishing the severe chronic pancreatitis from normal pancreas and mild pancreatitis, though there was no significant difference between mildly chronic pancreatitis and normal pancreas in metabolite concentration. The most representative metabolites were Aspartate, Betaine and Aspartate .HR - MAS NMR can be used as an effective method in the diagnosis and the rating of chronic pancreatitis.Objective:To explore the correlation of pathology (infiltration of inflammatory cells and fibrosis) and metabolites (Asp, Bet, FA) in rats .Methods: By the method of David J, on the basis of the degree gland atrophy, fibrosis, edema and inflammatory cell infiltration in pancreatic tissue(based on Partâ… ), chronic pancreatitis is rated for mild, moderate, and severe degree groups. With the data of integral area of major metabolites (Asp, Bet, FA), the correlation of pathology and metabolic was analysised by using Spearman rank correlation. Results : The correlation coefficient between Bet and fibrosis was 0.454(p=0.044), The correlation coefficient between Bet and infiltration of inflammatory cells was 0.716 (p=0.000);and FA and fibrosis were -0.768(p=0.000), -0.394(p=0.085),while the value of Asp , FA and infiltration of inflammatory cells -0.764 (p=0.000)and -0.619(p=0.004). Conclusion:There are correlation between pathology (fibrosis of chronic pancreatiis and fibrosis) and metabolites (Asp, Bet, FA) in rats. The correlation of Bet and infiltration of inflammatory cells and fibrosis of chronic pancreatitis was positive; the correlation of Bet and infiltration of inflammatory cells and fibrosis of chronic pancreatitis was negative, and Asp has negative correlation with fibrosis of chronic pancreatitis, while there were no correlation between Asp and fibrosis of chronic pancreatitis. |
