Objective 1.To investigate the feasibility of isolation, culture and identification of brain tumor stem cells and tumor cells in human glioblastoma.2.Aanlysis the exprrssion difference of ABCG2,SHH,EGFR,BCL2L12,MMP9 MMP2 gene between glioma stem cells and tumor cells.Methods 1.Isolate and culture brain tumor stem cells and tumor cells in human glioblatoma, reference neural stem cells in vitro culture condition. The tumor stem cells clone-spheres were identified with immunocytochemistery for CD133 and nestin.2.Trizol was used to isolation the RNA of tumor stem cells and tumor cells in human glioblatoma.3.Real-time quantitatie polymerase chain reaction (PCR) was adopted to Aanlysis the difference of ABCG2,SHH,EGFR,BCL2L12,MMP9,MMP2 gene between glioma stem cells and tumor cells.Results 1.CD 133 and nestin immunocytochemistery positive neurospheres can obtained from human glioblastoma clone-spheres after three days cultured.2.The PCR result suggest that ABCG2,SHH,EGFR,BCL2L12,MMP9,MMP2 in turner stem cells high expression compared with the expression in turner cells, Statistical analysis indicate that ABCG2 (t=4.87; P<0.001),SHH(t=7.68; P<0.001),EGFR(t=20.39; P<0.001),BCL2L12(t=5.79; P<0.001),MMP9(t=7.86; P<0.001),MMP2(t=7.31; P<0.001) between turner stem cells and turner cells。At the same time, the expression rate of ABCG2,SHH,EGFR,BCL2L12,MMP9,MMP2 are different. (F=4.18, F0.05.5.60,=2.37<4.18,P<0.05)。 Conclusion 1.Within human glioblastoma there were brain tumor stem cells, which has the same properties with normal neural stem cells. CD 133 and nestin are key surface markers for those cells.2.Brain tumor stem cells high expression ABCG2,SHH,EGFR,BCL2L12,MMP9,MMP2, this is quite important for the therapy of malignant glioma according to tumor stem cells.
