| Research Purpose:This paper is meant to find out the possible mechanism of acupuncture accelerating the healing of the skeletal muscle contusion by investigating the impacts that acupuncture has on the process and quality of the wounds' healing after rats suffer from skeletal muscle contusion,and discssing the impacts that the implemetation of acupuncture on SD (Sprague-Dawley) rats with skeletal muscle contusion has on pathological changes,and the changes of collagen fibers and PCNA(proliferating cell nuclear antigen)amount in different periods.Research Methods:After anesthetizing the rats by injecting 2% sodium(0.25ml/100g) into abdominal cavity,and performing blunt attack on the right hindlimb gastrocnemius muscle of rats (Kami, 1992), the researcher divides the rats into two groups randomly: one group for acupuncture treatment, the other group for natural healing. Kill some rats and take the wounded muscle the moment the rats are wounded. Kill some other rats and take the wounded muscle one day later,two days later,four days later,and six days later.After that,we can observe and analyze the pathological changes of its injureld muscle by using HE stain, analyze the collagen fibers' changes of injured muscle by using Mallory trichrome strain, and detect the contents of proliferating cell nuclear antigen by using immunohistochemistry.Research Results:(1)From the findings of HE stain,in the acupucture treatment group,the infiltration of inflammatory cells (mainly neutrophils, giant macrophage) in the injured partg is more concentrated and obvious in the first few days after injury, when inflammatory reaction is the most active,compared with the natural healing group.And the neonatal myotube of this group also appears earlier and the number is lager.Six days after injury, injure muscle fibers of the rats in this group arrang neatly and tightly. Besides,the inflammatory cells have dispersed. Meanwhile,there are no obvious scars formed, and the injuries have basically healed.However,the findings of the natural healing group are almost opposite.There is edema in the injured muscle fibers of the rats . In addition, muscle fibers in the injured part arranges loosly,and the structure of fibers in the amoeboid and necrotic parts hasn't recovered .What' more,there is still the infiltration of inflammatory cells and scars are being formed.(2) In both groups , there are muscle satellite cells appearing on the first day after injur. In the acupuncture treatment group, the number of muscle satellite PCNA-positive nuclei rises to its peak two days after injury, while the number of those in the other goup rises to its peak four days after injury, and in both groups there is a decrease in the number of them six days after injure. However, the number of them in the natural healing group is smaller than the number of muscle satellite PCNA-positive nuclei appearing on the first day, the second day andt he fourth day after injury.There is a significant difference(P<0.01) two days after injury.(3)After using acupuncture treatment, in the acupuncture treatment group, the amount of positive collagen fibers' formation in skeletal muscle repair tissues increases significantly six days after injury,while in the natural healing group it increases slowly. There is no obvious difference in the amount of collagen fibers' formation between the two groups within four days after injury, while the amountr of that in both groups is on the rise within six days after injury.Conclusion:(1) In the eary stage of acute skeletal muscle contusion, acuquncture will enable inflammation of the injuried part to appear early and subside in time, so that necrotic tissues of rats can be removed in a relatively short time. Acuquncture can accelerate the formation of cellagenous fibers and even ensure its stablity after rats suffer from acute skeletal muscle contusion(2)During the period of the restoration of acute skeletal muscle contusion ,acuquncture can accelerate the proliferation of skeletal muscle satelite clells of rats and advance its peak ;also can speed up the regeneration of muscle. (3)The implementation of acuquncture after rats suffer from acute skeletal muscle contusion can effectively speed up the regeneration of skeletal muscle in the injuried part and advance the process of tissues healing. |
