| Aim:To investigate the effects of rhizoma drynariae flavonoids on serum levels of leptin (LEP), interleukin-6(IL-6), prostaglandin E2 (PGE2) and the expression of boneβ2-adrenergic receptor (ADRB2), dopamine-(3-hydroxylase (DBH) in the rat model of ovariectomized osteoporosis (OP).Methods:1. Forty-five 3-month-old SD rats were randomly divided into model group (30) and sham operation group (15).The model group which are ovariectomized rats, after observation twelve weeks to determine whether the osteoporotic model was established successfully. Then the rats in the model group were divided uniformly into untreated model group(15) and rhizoma drynariae total flavones group(15), The rats in the rhizoma drynariae total flavones group were respectively treated with rhizoma drynariae total flavones,dose:50 mg·kg-1·d-1. And those in the sham-operated group and untreated model group received gastric perfusion of normal saline.Twelve weeks after treatment; Enzyme-linked immunosorbent assay was applied to detect the concentrations of LEP, IL-6 and PGE2 in serum. The expression of ADRB2 and DBH were determined by immunohistochemical technique.2. The osteoporotic model was established by ovariectomy.3. Twelve weeks after operation, the bone mineral density (BMD) was detected by dual X-ray energy to determine whether the osteoporotic model was established successfully.RESULTS:1. The BMD of rats in model group decreased significantly compared to sham group 12 weeks after operation (P<0.01), which indicated that the osteoporotic model was established successfully.2. The DBH expression in sham group and treatment group was significantly different from model group, and no significant difference between sham group and treatment group. The ADRB2 expression in sham group and treatment group was significantly different from model group, and no significant difference between sham group and treatment group.3. The serum levels of LEP, IL-6 and PGE2 in model group were significantly higher than those in sham group (P<0.05). The levels of LEP, IL-6 and PGE2 in rhizoma drynariae total flavones group were significantly lower than those in model group (P<0.01). No significant difference of PGE2 between groups was observed.CONCLUSION:1. The serum levels of LEP, IL-6 and PGE2 increase in OP rats, so as the expression of bone ADRB2 and DBH.2. Rhizoma drynariae total flavones reduce the production of LEP, IL-6 and expression of ADRB2, DBH; suppress the bone absorption, which may be the mechanisms of osteopractic total flavones in treating OP. |
