| Objective:To study the influence of glucose concentration on morphological characteristics and F-actin Cytoskeleton changing in cultures of rabbit corneal endothelial cells in vitro.Methods:Rabbit cornear endothelial cells were cultured in different glucose concentrations and observed by phasecontrast microscopy, flowcytometry, transmission electron microscopy(TEM),laser confocal seanning microscopy(LSM) and atomic force microscopy(AFM) the morphological characteristics and F-actin Cytoskeleton changing.Results:By the concentrate of the glucose elvated, the corneal endothelial cells had changed in morphology, such as swelling of cells, a decreased percentage of hexagonal cells.The apoptosis cells increased and the content of F-actin had induced.TEM shows that the mitochondria exhibited vacuole degeneration and the crista broke and the rough endoplasmic reticula expanded. LSM shows that the cytoskeleton proten had changed,AFM shows that membrane ultrastructur had changed and the elasticity had increasedConelusion:These results indicated that the elevated glucose influenced the structure of corneal endothelial cells and the reserved function of corneal endothelial cells were abnormal. |
