| The radix of Euphorbiae Ebracteolatae was used in traditional Chinese medicine for softening hardness to Dissipating stagnation, relieving oedema, eliminating pains and anti-worms. Euphorbiae Ebracteolatae Radix contains diterpenes, acetophenone, triterpenes, flavonoids, sterols and other components. Diterpenes and acetophenone are the characteristic components in it, the typical active component.In order to exploit and utilize the resources of Euphorbiae Ebracteolatae Radix, and establish the quality standards, this article firstly make systematically analyzing on the chemical constituents of the petroleum ether extract parts, which is from 95% ethanol extract of Euphorbia ebracteolata Hayata.10 compounds had been separated through various chromatographic techniques, the structure of which was identified by 1HNMR,13CNMR,HMQC,HMBC and MS spectral analysis and referring to literatures data. They were P-amyrin acetate (1),24-Methylenecycloartanol (2),2,4-Dihydroxy-6-methoxy-3-methylacetophenone (3),β-sitosterol (4), yuexiandajisu D (5),2,2',4,4'-tetrahydroxy-6,6'-dimethoxy-3,3'-dimethyl-7,5'-bisacetophenone (6), Jolkinolide B (7),2,4-dihydroxy-3-formyl-6-methoxy-acetophenone (8),3,3'-diacetyl-4,4'-dimethoxy-2,2',6,6'-tetrahydroxy-diphenylmethane (9) and Jolkinolide A (10). A new compound Jolkimethyl ester (11) was discovered during the study of the instability of Euphorbiae Ebracteolatae in methanol solven. This will be important in the selection of the solvent during the determination of Euphorbiae Ebracteolatae Radix.On the basis of the study of chemical composition, this article firstly established the method for determination of 2,4-Dihydroxy-6-methoxy-3-methylacetophenone and Jolkinolide B in Euphorbiae Ebracteolatae Radix, by measuring the 2,4-Dihydroxy-6-methoxy-3-methylacetophenone and Jolkinolide B in collected Euphorbiae Ebracteolatae Radix, and make some recommended limits for the reference of Chinese Pharmacopoeia. Meanwhile, it is the first time to establish the fingerprint of Euphorbiae Ebracteolatae Radix, providing theoretical basis for the identification of Euphorbiae Ebracteolatae Radix. In this article, Bioactive Constituents of Euphorbiae Ebracteolatae Radix are analyzed by using the high performance liquid chromatography-electrospray ionization-quadrupole-time of flight mass spectrometer. Mass spectrometry cracking rule for Diterpenes and acetophenone, the major elements of Euphorbiae Ebracteolatae Radix, was summarized through level 1 mass spectrometry and second- order mass spectrum, and with review of the literature. It can detect the chemical components of Euphorbiae Ebracteolatae Radix in a rapid and comprehensive way, and also provide an effective basis for its quality control. Finally, a full quality control standard for Euphorbiae Ebracteolatae Radix was established base on the above researches. |
