| Objective To investigate the effects of local application of NGF combine with Insulin on wound healing with II degree deep scald and the expression of HIF-la and VEGF in diabetic rats Methods 60 male Wistar rats were used in the study by intraperitoneal injection of streptozotocin (STZ). Deep partial thickness scalding was engendered on the back of the rats after one month. Then these rats were randomly divided into B (scalding of the rats with diabetes,n=15), C (insulin control, n=15), D (NGF control, n=15) and E (NGF and insulin supplementation,n=15)groups, and group A (nomal control,n=15) was created by normal rat of partial thickness scalding, The wound area, wound healing rate, and grey of HIF-la and VEGF immunohistochemistry staining were determined on 3,7,15, and 21 day post scalding days (PSDs). Results Compared to those in groups A,B,C and D, the wound healing rate in group E (25.33±2.32)% increased significantly since the 7th PSD (P<0.05). The wound healing rate was the highest in group E (39.74±3.22)% and the lowest in group B (23.60±2.27)% compared to other groups with significantly different, (P<0.05) on 11th day. There were no significant difference in wound healing rate between group A (34.29±2.69)% and D (32.72±2.37)%(P>0.05). The wound healing rate was had a cross between group A and group D on about 13th day, and after that the wound healing rate in group D (54.84±3.60)% began to surpassed group A (50.40±3.01)% from 15th day and there was significant difference between them (P<0.05). Till 21st day, the wound healing rate was the highest in group E (83.62±5.56)% and the lowest in group B (56.26±4.28)% compared to other groups with significantly different (P<0.05). Compared to group A (75.65±4.52)%, the group D (78.03±4.49)% increased significantly on the 21st PSD (P<0.05). Group C (71.97±4.19)% was in between group D and group A. The expression of HIF-1α(92.77±6.63) and VEGF (110.73±4.92) in group E began to show lower than other groups from 7th day till 21st day(P<0.05). The expression of HIF-1αand VEGF in each group began to decrease after they reach to peak on 15th day. The grey of HIF-la and VEGF were had a cross between group A and group D on about 13th day, and after that the expression of HIF-1αand VEGF in group D began to less than group A. Till 21st day, the expression of HIF-la (117.78±8.42) and VEGF (123.40±3.40) were lowest in group E compared to other groups with significantly different, (P<0.05). Conclusion Local application of NGF combine with Insulin could be beneficial to the regeneration of capillary vessel in the burn wounds of the rats with diabetes, as well as to accelerate wound healing by redress anoxia and decreasing the expression of HIF-1αand VEGF. |
