【Objective】Studied the protective effect of phosphocreatine in myocardial injury caused by spinal cord injury and possible mechanisms.【Methods】54 healthy male SD rats, weighing 250 ~ 300g,were randomly divided into nine groups, n = 6.:the control group with operation (TC), injury group (TS): SCI injury 6h (TS1), 12h (TS2), 24h (TS3) and 48h (TS4) and intervention group (TL): SCI injury 6h (TL1) , 12h (TL2), 24h (TL3) and 48h (TL4) injecting phosphocreatine into SD rat abdominal (200mg/kg),harvesting in 2h after injection.Them were harvested from SD rats abdominal aorta with scalp needle, collecting the blood 2ml, measured troponin I(TnI)levels and the creatine kinase (CK), creatine kinase isoenzyme (CK-MB) levels and other biochemical laboratory enzyme.Cutting SD rat cardiac apex organization extracted elevant materia with protein lysate cracking .Using fluorescence quantitative PCR determined the CasR mRNA expression and using Western blot determined the CaSR protein in myocardial tissue. using the transmission electron microscope to observed morphological features of myocardial cell injury.【Results】The cTnI and CK, CK-MB levels of TS,TL groups at each time point were higher than TC group (P <0.05); cTnI level of TS group peaked at T1 and T2, T3, T4 time points followed by decreasing level at T4 and was still lower than the control group (P <0.05); CK level of TS group peaked at T3, the level at T4 was still lower than the control group (P <0.05); CK-MB level of TS group peaked at T2, T4 point level was still lower than the control group (P <0.05); In TL group, the contents of TnI and CK, CK-MB levels than the corresponding point in each TS groups wear decreased (P <0.05). The expression of the CasRmRNA at each time point in TS groups and TL groups were higher than the TC group (P <0.05); The CasRmRNA expression in TL group were lower than the TS group at the same time point (P <0.05); The CasRmRNA expression of TS group at T1, T2, T3 increased gradually, T4 time point lower than the other but still higher than TC group (P <0.05); When TL compared with the TS, the most obvious difference of the CasRmRNA expression at each time was visible at T3 The expression of the CasRmRNA at each time point in TS groups and TL groups were higher than the TC group (P <0.05); The CasR protein expression in TL group were lower than the TS group at the same time point (P <0.05); The CasR protein expression of TS group at T1, T2, T3 increased gradually, T4 time point lower than the other but still higher than TC group (P <0.05); When TL compared with the TS, the most obvious difference of the CasR protein expression at each time was visible at T3 .ITEM results show that most of the structure of myocardial cell nucleus,sarcomere are integrity and muscle wire is clear in TL group;the structure of mitochondria is normal,no swelling and vacuolar degeneration,the ridge structure is clear ;the myocardial damage is significantly Light than TS group at corresponding time points.【Conclusion】Spinal cord injury in rats can cause the diversification of myocardial enzyme, morphology and CasR highly expression and not to return to normal in 48h..Phosphocreatine can improve myocardial enzyme, morphology and other aspects in Spinal cord injury rats .Phosphocreatine can reduce the expression of CasR in Spinal cord injury rats, and the most significantly in the 24h, continued until 48h.
