| Objects: To explore the mechanism of San miao hong teng decoction on the Treating Menorrhagia Damp-heat Syndrome, Supply experiment basis for its clinical effect.Methods:50 adult female SD rats which were not pregnant, selec t 42 of which modeling injected with Escherichia coli,the remaining 8 rats were divided into normal control group。The model rats were ran domly divided into four groups after 14 days:San miao hong teng deco ction higer dosage group, San miao hong teng decoction lower dosage g roup, gong xue ning group, model control group. Medicine were continua lly given to the rats in fourteen days, then select six rats in each groups, take of arterial blood and check blood testof PT, APTT, PLT; Take uterus organization, detect the number of inflammatory cells by hematoxylin-eosin stainingdyeing(HE)and TNF-a, NF-kB, tPA, TIMP-1, MMP-2 by immunohistochemistry.Result:1. the number of inflammatory cells:Compared with the normal control group, the number of model control group were increased (p<0.05).Compared with the model team, the number of San miao hong teng decoction higer dosage group, lower dosage group and gong xue ning group are significantly reduced (p<0.05). The function of san miao hong teng decoction higer dosage group reduced inflammatory cells are higher than gong xue ning group, but between san miao hong teng decoction lower dosage group and gong xue ning group, between San miao hong teng decoction higher dosage and lower dosage, the groups compared have no remarkable difference(P>0.05).2. Hemostatic:Compared with the normal control group, the model team rat's PLT are reduced (P<0.05), rat's PT and APTT are increased (P <0.05). Compared with the model team, San miao hong teng decoction higer dosage group, lower dosage group, and gong xue ning group can increase rat's PLT (P<0.05), reduce rat's PT and APTT (P<0.05). San miao hong teng decoction higer dosage group rising rat's PLT is lower than gong xue ning group (P<0.05). Between San miao hongteng decoction higer dosage group and San miao hong decoction lower dosage group, between San miao hong decoction lower dosage group and gong xue ning group, groups comparions rat's PLT have no significant difference (P>0.05). San miao hong teng decoction higher dosage group, San miao hong teng decoction lower dosage group and gong xue ning group, between them the groups comparions rat's PT and APTT have no significant difference(P>0.05).3. Endometrial TNF-α, NF-KB, tPA, TIMP-1, MMP-2 expression:Compared with the normal control group, the model team's endometrial TNF-α, NF-kB , tPA, MMP-2 expression are increased (P<0.05), endometrial TIMP-1 expression are reduced (P<0.05). Compared with the model team, San miao hong teng decoction higer dosage group, lower dosage group and gong xue ning group can reduce endometrial TNF-a, NF-kB, tPA, MMP-2expression ( P<0.05), increase endometrial TIMP-1 expression (P<0.05). San miao hong teng decoction higher dosage group, San miao hong teng decoction lower dosage group and gong xue ninggroup, between the groups endometrial TNF-α, MMP-2expression have no significant difference (P>0.05). San miao hong teng decoction lower dosage group rising tPA expression is higher than gong xue ning group (P<0.05). Between San miao hong decoction higer dosage group and gong xue ning group, between San miao hong teng decoction higer dosage group and San miao hong decoction lower dosage group, tPA expression has no significant difference (P>0.05). San miao hong teng decoction higer dosage group and San miao hong decoction lower dosage group NF-kB expression are lower than gong xue ning group (P<0.05) Between San miao hong teng decoction higer dosage group and San miao hong decoction lower dosage group, NF-kB expression has no significant difference (P>0.05). San miao hong teng decoction lower dosage group rising endometrial TIMP-1 expression is higher than San miao hong teng decoction higher dosage group and gong xue ning group (P<0.05). Between San miao hong teng decoction higher dosage group and gong xue ning group , TIMP-1 expression has no significant difference (P>0.05).Conclusions:(1) Escherichia coli endometrial inoculated can effectively be used to establish the model rats with pelvic inflammation Disease. (2) Experimental studies suggest San miao hong teng decoction can increase PLT and shorten PT, APTT. Affect the function of blood coagulation and hemostasis; San miao hong teng decoction can reduce inflammatory cells, inflammatory factor TNF-α, NF-kB, tPA, inhibite the degradation of xtracellular matrixc(ECM), reduce endometria's off. then San miao hong teng decoction can anti-inflammatory and hemostatis. (3) San miao hong teng decoction higher dosage group and San miao hong teng decoction lower dosage group have no dose-effect relationship. |
