Objective:To take TCM and acupuncture theory as the guide, using the methods of ethology and molecular biology to explore the effect of the hypothalamic CRH for electro-acupuncture's regulative action of chronic fatigue syndrome rats.Methods:Health experimental SD rats were randomly divided into normol control group,model group,mould electro-acupuncture (EA) group,mould false injection group,mould CRH injection group,mould CRH antagonist injection group,mould CRH injection EA group and mould CRH antagonist injection EA group. Using the methods of chronic restraint and cold water swimming to make the chronic fatigue model. Using the test of exhaustion swimming,rat tail suspension and water maze to evaluation after model making. Implement lateral ventricle injection for mould false injection group,mould CRH injection group,mould CRH antagonist injection group,mould CRH injection EA group and mould CRH antagonist injection EA group after model evaluation.The next day, mould EA group,mould CRH injection EA group and mould CRH antagonist injection EA group with electro-acupuncture on shenshu and zusanli.To detect and analysis the ethology and plasma corticosterone(CORT) after treatment.Results:①The results of exhaustive swimming test:Each group's exhaustive swimming times were significant statistical differences(P<0.05). Mould CR H injection group compared with normol control group, there were significa nt differences(P<0.0018);The following group compared with each other, the re were no significant statistical differences(P>0.0018). But model group compared with normol control group has significantly trend to shorten, mod el EA group compared with model group has significantly trend to prolong, mould false injection group compared with model group has no significantl y trend to shorten,mould CRH injection group and mould CRH antagonist inj ection group compared with model group have significantly trend to shorte n,mould CRH injection group compared with mould CRH antagonist injection group has significantly trend to prolong, mould CRH injection EA group com pared with mould CRH injection group has significantly trend to prolong,m ould CRH antagonist injection EA group compared with mould CRH antagonist injection group has significantly trend to prolong.②The results of rat tail suspension test:Each group's exhaustive swim ming times were significant statistical differences(P<0.05). Mould grope,mould CRH injection group and mould CRH injection EA gr oup compared with normol control group, there were significant differences (P<0.0018);The following group compared with each other, there were no sig nificant statistical differences (P>0.0018). But model EA group compared wi th model group has significantly trend to prolong, mould CRH injection EA group compared with mould CRH injection group has significantly trend to prolong, mould CRH antagonist injection EA group compared with mould CRH a ntagonist injection group has significantly trend to prolong.④Blood plasma CORT content:Each group's exhaustive swimming times wer e significant statistical differences (P<0.05). Mould group compared with n ormol control group has increased,there were significant differences(P<0. 05),mould EA group compared with mould group has decreased,there were sig nificant differences(P<0.05). The following group compared with each other, there were no significant statistical differences(P>0.05). But mould CRH i njection group and mould CRH antagonist injection group compared with mod el group have significantly trend to decrease, mould CRH injection group c ompared with mould CRH antagonist injection group has significantly trend to increase, mould CRH injection EA group compared with mould CRH injecti on group has significantly trend to increase, mould CRH antagonist injecti on EA group compared with mould CRH antagonist injection group has signif icantly trend to increase.Conclusions:Hypothalamic CRH secrete abnormal lead to chronic fatigue. Electro-acupuncture on shenshu and zusanli adjusting hypothalamic CRH content and plasma CORT, so as to improve HPAA function and relieve fatigue.
